比色法测定比格犬尿液中柠檬酸的浓度

摘　　要：建立了一种测定比格(Beagle)犬尿液中柠檬酸浓度的方法,利用柠檬酸与溴化钾、高锰酸钾在酸性溶液中生成五溴丙酮,五溴丙酮经石油醚(90℃~120℃)萃取后,与硫脲反应生成水溶性黄色复合物,其用紫外分光光度计在435nm波长处测定。结果显示,柠檬酸浓度在0.005mg/mL^0.25mg/mL范围内,水溶性黄色复合物的吸光度与柠檬酸的浓度呈良好的线性关系,OD435=1.8335C+0.0038,r=0.9996,平均回收率为99.954%(n=5),变异系数为1.64%(n=20)。该方法操作简单,快速准确,适用于非临床研究中Beagle犬尿液中柠檬酸浓度的测定。The purpose of this study was to establish a method for measuring citric acid in urine of Beagle and evaluate application in non-clinical research.In this study, citric acid, bromatum kalium and potassium permanganate can act and generate pentabromoacetone in acid solution.The pentabromoacetone was extracted by petroleum ether and then reacted with thiourea to generate yellow compound of water solubility.And it can be measured by spectrophotometer in 435 nm wavelength.The final results in treated urine were determinated exactly.All the coefficients of variation were less than 5 %.The recoveries for the five urine samples were all more than 95%.The methods is simple,accurate and fast.It is suitable for the determination of citric acid in the urine of Beagle.
