摘  要:目的分析云南省手足口病(hand,foot and mouth disease,HFMD)患儿标本中柯萨奇病毒A10型(coxsackievirus A10,CV-A10)病毒VP1区基因特征。方法采集2012-2014年云南省HFMD患者粪便和咽拭子标本,用细胞培养法分离病毒,提取病毒RNA,用RT-PCR法进行病毒VP1区基因序列测定,用基因分型方法鉴定病毒的血清型和基因型别。按文献选择CV-A10各基因型的代表株,用MEGA 5.0软件构建CV-A10基因进化树并分析核苷酸和氨基酸的同源性。结果 2012-2014年云南省共采集到21 761份HFMD病例的临床标本,对其中的2 578份标本进行病毒分离,共分离到608株病毒,其中17株为CV-A10。它们之间的核苷酸(nucleotide,nt)和氨基酸(amino acid,aa)同源性分别为94.30%~100.00%和98.04%~100.00%,与原型株(Kowalik,AF081300)的nt和aa同源性为76.23%~76.71%和91.18%~92.65%。基因进化分析表明CV-A10云南分离株为C基因型,它们又分为两个进化分支。结论CV-A10是除EV-A71和CV-A16外引起手足口病的重要病原之一,和国内报道一样,云南省流行的CV-A10为C基因型,2012-2014年有两个进化分支在云南共同流行。Objective To analyze the genetic features of VP1 gene of coxsackievirus A10 (CV-A10) strains iso- lated from hand, foot and mouth disease (HFMD) patients in Yunnan province of China from 2012 to 2014. Methods Stool and nasopharyngeal swab samples were collected from HFMD patients from 2012 to 2014. Viruses were isolated by cell culture. Virus RNA was extracted, the VP1 gene was amplified by reverse transcription-polymerase chain reaction (RT-PCR) and sequenced. The serotype was identified by molecular typing method and the genotyping was determined by phylogenetic analysis. Results A total of 21 761 clini- cal samples were collected from HFMD patients during 2012 to 2014, virus isolation was done from 2 578 sam- pies and 608 virus strains were isolated and of which, 17 strains were CV-A10. The homology of nucleotide (nt) and amino acid (aa) of the 17 CV-A10 strains were 94.30%-100.00% and 98.04%-100.00%, respectively; these CV-A10 strains share 76.23%-76.71% and 91.18%-92.65% in homology with the prototype strain (Kowalik, AF081300). Phylogenetic analysis showed that all Yunnan CV-A10 strains were grouped to genotype C. Conclusions Genotype C coxsackievirus was the main circulating strain in Yunnan province of China from 2012 to 2014.
