中药养阴通便方调控肠道β-arrestin2介导的信号通路改善癌症患者便秘的机制研究

摘　　要：目的分析中药养阴通便方通过调控肠道β-arrestin2介导的信号通路对癌症患者便秘情况的影响。方法选取50例癌症便秘患者,随机分为对照组和试验组,对照组不采取任何便秘治疗措施,试验组服用中药养阴通便方。记录患者治疗前以及治疗后1个月患者生活质量自评量表(PAC-QOL)评分及Cleveland便秘评分。观察两组β-arrestin2介导的信号通路相关基因的表达情况。结果治疗前对照组与试验组各观察指标比较无显著差异(P>0.05)。对照组治疗前后PAC-QOL评分及Cleveland便秘评分无显著差异(P>0.05),试验组治疗后PAC-QOL评分及Cleveland便秘评分较治疗前均明显下降,且明显低于对照组,差异均有显著性(P<0.05)。试验组治疗便秘总有效率(84.00%)明显高于对照组(4.00%),差异有显著性(P<0.05)。试验组β-arrestin2介导的信号通路相关基因的相对表达量,除了Akt低于对照组外,PP2Ab、PP2Ac、m TOR均高于对照组(P<0.05)。结论中药养阴通便方可以通过调控β-arrestin2,促进癌症便秘患者的肠道蠕动,加速患者排便,提高患者生活质量。Objective To investigate the effect of traditional Chinese medicine Yang - Yin - Tong - Bian - Recipe on the constipation of cancer patients by regulating β- arrestin2 - mediated signaling pathway. Methods Fifty tumor patients with constipation were randomly divided into the control group and the experimental group. The control group did not exert any influence, and the patients in the test group were treated with the recipe. QOL score and Cleveland score of patients with constipation were recorded before and 1 month after treatment. The expression of β- arrestin2 mediated signal transduction related genes in two groups was observed. Results There were no significant differences between the control group and the experimental group before treatment （ P 〉 0. 05 ） . There was no significant difference in the PAC - QOL score and the Cleveland score between the two groups （ P 〉 0. 05） , after treatment, the PAC - QOL score and Cleveland score of the experimental group were significantly lower than those before the treatment, and significantly lower than those in the control group, the difference was significant （ P 〈 0. 05）. The total effective rate （84. 00% ） in the treatment group was higher than that in the control group （4. 00% ） （ P 〈0. 0 5 ） . Compared with the control group, PP2Ab, PP2Ac and mTOR were significantly higher than those in the control group （ P 〈 0 .0 5 ） ,and excepting the gene Akt, the relative expression of beta - arrestin2 mediated signaling pathway in the experimental group was higher than that in the control group. Conclusion Traditional Chinese medicine Yang - yin - Tong - bian Recipe can regulate p - arrestin2 to promote cancer patients with constipation intestinal peristalsis, accelerate cancer patients with constipation defecation and improve the quality of life of patients.
