摘  要:为研究猪瘟(CSF)、猪繁殖与呼吸综合征(PRRS)免疫猪肺门淋巴结中淋巴细胞对PRRSV N类特异性肽刺激的应答特点,试验从猪肺门淋巴结中分离T淋巴细胞,分别用PRRSV N类特异性肽进行刺激,待细胞培养至24,48h时,离心取细胞上清液,检测上清液中细胞因子TGF-β1、IL-10、IL-12分泌含量的变化。结果显示,PRRSV N类特异性肽能够抑制TGF-β1、IL-10的分泌,表明N1、N2肽具有减弱免疫抑制,促进抗炎性细胞因子分泌的作用;此外IL-12的分泌量也减少,且有逐渐恢复平衡的趋势,提示PRRSV N类特异性肽可能具有双面性,有待于进一步研究。本试验为PRRSV合成肽疫苗及疫苗佐剂的研制提供理论依据。In order to analyze that response characteristics of lymphocytes in hilar lymph nodes of pigs immuned by classical swine fever (CSF)and porcine reproductive and respiratory syndrome (PRRS)vaccine under the stimulation of PRRSV N class specific peptides. In this study, T lym- phocytes from porcine hilar lymph nodes were isolated and stimulated with PRRSV N class specif- ic peptides. At 24 and 48 h after stimulated,the supernatant liquid after centrifugation was used to test the secretion of TGF-β1 ,IL-10 and IL-12. The results showed that the PRRSV N class specific peptides could inhibit the secretion of TGF-β1 and IL-10,indicating that the specific peptides could weak the immunosuppression and promote the then showing anti-inflammatory cytokine produc- tion. In addition, the secretion of IL-12 decreased and then showing a tendency to gradually restore balance,indicating that PRRSV N class specific peptides may have a double effect,which awaits a further discussion. This study provided data for the development of PRRSV synthetic peptidevaccine and vaccine adjuvant.
