摘  要:目的:探讨巢蛋白对心肌梗死后大鼠缺血心肌再修复的影响。方法:将雄性Sprague-Dawley大鼠30只随机分为对照组、心肌梗死损伤组及心肌梗死损伤修复组,每组各10只;心脏超声检测左室舒张末内径(LVEDD)、左室收缩末内径(LVESD)、左室射血分数(LVEF)、左室缩短分数(FS)。HE染色光镜下观察大鼠心肌形态学改变。Western检测心肌细胞巢蛋白、NKX2.5、GATA4表达。结果:心梗损伤组和心梗修复组均可见心肌细胞肥大,心梗损伤组还出现部分心肌细胞坏死,与心梗损伤组相比,心梗修复组大鼠心肌病理形态、左室大小、射血分数明显改善,巢蛋白、NKX2.5、GATA4表达明显增加(P<0.01)。结论:心肌发生缺血损伤后巢蛋白的表达会增加,巢蛋白参与了损伤心肌的再修复,对损伤心肌具有修复作用。Objective: To explore the effect of nestin on the repairment of the damage of ischemic myocardial. Methods: 30 male SD rats were randomly divided into control group, myocardial infarction group and myocardial infarction damage repairment group (10 for each). Left ventricular end diastolic diameter (LVEDD), left ventricular end systolic diameter (LVESD), left ventricular ejection fraction (LVEF), left ventricular shortening fraction (FS) were measured by echo- cardiography HE staining was used to observe the morphological changes of myocardium in rats. Western was used to de- tect the expression of nestin, NKX2.5 and GATA4 in myocardial cells. Result: Cardiomyocardial hypertrophy was ob- served in myocardial infarction group and myocardial infarction group. There were some myocardial necrosis in myocardial infarction group. Compared with the myocardial infarction group, the myocardial pathological morphology, left ventricular size and ejection fraction were significantly improved; the expression of nestin, NKX2.5 and GATA4 were significantly increased in the myocardial infarction damage repairment group ( P 〈 0.01 ). Conclusion: The expression of nestin in- creases after myocardial ischemia injury. Nestin is involved in the repairment of damaged myocardium, and it can repair the injured myocardium.
