摘 要:利用逆转座子间扩增多态性(Inter-retrotransposon Amplified Polymorphism,IRAP)分子标记对收集的49份江西柿属种质资源进行了遗传多样性分析。从26个IRAP引物中筛选出扩增条带清晰且多态性丰富的10个引物,PCR扩增产物经聚丙烯酰胺凝胶电泳(PAGE)得到清晰的指纹图谱。将418条清晰条带(多态性100%)转换为0/1矩阵,聚类分析结果表明:供试的江西柿属种质资源的遗传相似系数在0.69~0.92之间,分属柿、油柿、野柿三个种,遗传多样性丰富;明晰了几个分类关系模糊的柿种质资源的种属关系。The genetic diversities of 49 germplasm resources in Diospyros in Jiangxi were analyzed by using IRAP ( Inter-ret- rotransposon Amplified Polymorphism) molecular marker. Ten primers which had clear bands and polymorphism in PCR amplifi- cation were screened out from 26 IRAP primers, and the PCR-amplified products presented clear fingerprint by polyacrylamide gel electrophoresis (PAGE). The 418 clear bands (100% diversity) were transformed into 0/1 matrix, and the clustering analysis showed that the genetic similarity coefficients among the tested 49 germplasm resources varied from 0.69 to 0.92, and these germplasm resources belonged to 3 species ( Diospyros kaki Thunb., Diospyros oleifera Cheng, and Diospyros kaki var. silves- tis Makino), which revealed abundant genetic diversity. In addition, the species taxonomic relationships among several Diospyros spp. germplasm resources were clarified.
