摘  要:通过水稻中OsWRKY45基因在NCBI中进行同源序列比对,电子克隆ZmWRKY45基因并与其他物种中的同源基因进行进化树分析。利用该基因的CDS区设计特异引物,在3叶期对吉单441进行不同浓度NaHCO_3处理,对玉米杂交种吉单441及其双亲自交系进行验证。结果表明,在吉单441及其父本中含有ZmWRKY45,在盐碱胁迫下吉单441中该基因随着盐碱浓度的升高而表达量升高,初步判断该基因可能在吉单441的耐盐碱过程中起到正向调控作用。By homologous sequence comparison of OsWRKY45 genes in NCBI, evolutionary tree analysis of electronic cloning ZmWRKY45 gene and homologous genes from other species, design of special primers according to the CDS area of the gene, maize hybrid Jidan441 and their parents inbred lines were respectively verified. In trilobites period, Jidan441 was treated by NaHCO3 with different concentrations. It showed that ZmWRKY45 exsist in Jidan441 and its male parent gene. In saline-alkali stressed Jidan441, gene expression could be improved with the in- creasing concentration of saline-alkali, which indicates that Jidan441 may have positive regulation effect in saline- alkali tolerant process.
