摘 要:目的:研究加味苓桂术甘方对酒精性脂肪肝大鼠肝脏组织脂肪分化相关蛋白(ADRP)、过氧化物酶体增殖活化物受体γ(PPAR-γ)和沉默信息调控子2(Sir2)蛋白表达的影响。方法:SD雄性大鼠60只,随机分为6组,每组10只。采用白酒灌胃,同时饲喂高脂饲料,制备酒精性脂肪肝模型。实验组动物给予不同剂量的加味苓桂术甘方灌胃6周。采用生化分析仪检测血清总胆固醇(CHOL)、甘油三酯(TG)、脂肪酸(FFA)的含量;利用实时荧光定量聚合酶链式反应(RT-PCR)和免疫印迹(Western blot)技术,检测大鼠肝脏组织中ADRP、PPAR-γ和Sir2的m RNA和蛋白表达。结果:与模型组比较,给药6周后加味苓桂术甘方中剂量组和大剂量组大鼠血清中TG、CHOL和FFA水平均明显下降(P<0.05);加味苓桂术甘方可显著下调肝脏组织中ADRP、PPAR-γ的表达,而上调Sir 2的表达。结论:加味苓桂术甘方可通过改变肝脏组织ADRP、PPAR-γ和Sir2的表达,从而抑制脂肪肝的发生。Objective: To study the effect of JWLGZG Decoction on expressions of ADRP, PPAR-γand Sir 2 protein in liver tissues in alcoholic fatty liver rat model. Methods: The 60 male Spague-Dawlayrats were randomly divided into 6 groups with 10 rats in each group. The rats were fed with high fat diet and ethanol fed to establish the experimental model of alcoholic fatty liver. The animals of experimental groups were treated with different doses of JWLGZG. The total cholesterol (CHOL), triglyceride (TG) and fatty acid (FFA) in blood of animals were measured using biochemical analyzer. The expressions of ADRP, PPAR-γ and Sir2 mRNA and proteins in liver tissues were analyzed through RT-PCR and western blot. Results: The levels CHOL, TG and FFA in serum of rat models treated with JWLGZG with medium-dosage and the high one were significantly lower than those of control group. JWLGZG could significantly decrease the expression of ADRP, PPAR-γ and up-regulated the expression of Sir 2 liver tissue. Conclusion: JWLGZG can change the liver tissue ADRP, PPAR-γ, and Sir 2 expression, thereby inhibit the occurrence of fatty liver.
