低机械指数超声介导声诺维对体内外细胞活性影响的研究

摘  要:目的:探讨低机械指数超声介导声诺维对人脐静脉内皮细胞和脑缺血模型大鼠脑细胞活性影响的研究。方法:体外实验:分为空白对照组(a组);单纯微泡剂组(b组);超声微泡组(c组),应用MTT测线粒体活性。体内实验:A组为单纯超声辐照组,B组为高剂量造影剂加超声辐照组,C、D、E组分别为低、中、高剂量造影剂加脑缺血造模超声辐照组。TUNEL法检测脑细胞凋亡情况。结果:体外实验显示,超声联合微泡剂(c组)对内皮细胞活性的影响有显著意义(P<0.001),但c组间差异无统计学意义。体内实验显示,凋亡细胞计数,C、D、E组与A、B组间有显著统计学差异(P<0.01),但在缺血C、D、E组间以及A、B组间无统计学差异(P>0.05)。结论:低机械指数超声造影可增强细胞通透性,但在控制好各种因素的前提下可安全应用于临床超声。Objective:to study the sound low mechanical index ultrasound mediated sonovue on human umbilical vein endothelial cells and cerebral ischemia model rats research on the effects of cell activity;Methods:in vitro experiment:cultivate human umbilical vein endothelial cells in vitro,divided into blank control group ( group a );Simple microbubble agent group ( group b ) , is divided into four levels(50mg/mL,25mg/mL,17mg/mL,12. 5mg/mL). Ultrasound microbubble group(c group),like group b is divided into four levels,as determined by MTT test application activity of the mitochondria. In vivo experiment:making acoustic window of Wistar rat model, group A is simple ultrasound irradiation group,group B contrast agent for high doses of ultrasound irradiation group,C,D,E group respectively for low,medium and high dose of contrast agents and cerebral ischemia mode ultrasound ir-radiation group. After the experiment,put to death in brain tissue,TUNEL method to detect the brain cell apoptosis. Results:the vitro experiments showed that the micro bubble group( group b) on vascular endothelial cell injury has no significant(P〉0. 05). Joint ultrasound microbubble agent(group c)effect on the activity of endothelial cells have a significant(P〈0. 001),but with the increase of microbubble agent concentration, reduce the cell survival rate did not see, there was no statistically significant difference between the group c. In vivo experiments have shown that in group A and group B there is randomly dispersed in the telencephalon,diencephalon apoptotic cells. A large number of apoptotic cells mainly distributed in ischemic necrosis around the stove in group C、D、E, the so-called &quot;half dark band&quot; area. Apoptotic cell count,C、D、E group had statistically significant difference between group A、B(P〈0. 01),but in ischemia-between group C、D、E and between group A,B has no statistical difference between groups ( P〉0 . 05 ) .
