摘 要:目的探讨p38丝裂原活化蛋白激酶(p38MAPK)特异性抑制剂SB203580对糖尿病小鼠肾小球滤过率的影响及机制。方法60只清洁级小鼠随机分为正常对照组(NG组)、糖尿病组(DM组)和SB203580治疗组(SB组)。测定小鼠血清超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量、尿白蛋白排泄和肾小球滤过率。结果与NG组比较,DM组血清SOD活性明显降低,MDA含量和尿白蛋白排泄明显增加(P〈0.05);与DM组比较,SB组血清SOD活性升高,MDA含量和尿白蛋白排泄降低(P〈0.05)。与NG组比较,DM组肾小球滤过率降低(P〈0.05);与DM组比较,SB组肾小球滤过率升高(P〈0.05)。相关性分析:肾小球滤过率与血清SOD活性存在正相关关系(r=0.670,P〈0.001),与血清MDA含量存在负相关关系(r=-0.731,P〈0.001)。结论p38MAPK特异性抑制剂SB203580通过减轻氧化应激改善肾小球滤过率。Objective To observe the effect of p38 mitogen-activated protein kinase ( p38MAPK ) specific inhibitor SB203580 on glomerular filtration rate in diabetic mice. Methods Sixty clean mice were randomly divided into normal control group ( NG group ) , diabetic group ( DM group ) and SB203580 treatment group ( SB group ) . The activity of superoxide dismutase ( SOD ) , content of malondialdehyde ( MDA ) , urinary albumin excretion and glomerular filtration rate were measured. Results ( 1 ) Compared with NG group, the activity of SOD in DM group was significantly decreased, the content of MDA and urinary albumin excretion were significantly increased ( P〈0.05 ) . Compared with DM group, the activity of SOD in SB group was increased, the content of MDA and urinary albumin excretion were decreased ( P〈0.05 ) . ( 2 ) The glomerular filtration rate of DM group was lower than that of NG group ( P〈0.05 ) , and the glomerular filtration rate of SB group was higher than that of DM group ( P〈0.05 ) . ( 3 ) Correlation analysis: The glomerular filtration rate was positively correlated with the activity of SOD ( r=0.67, P〈0.001 ) , and the glomerular filtration rate was negatively correlated with the content of MDA ( r=-0.731, P〈0.001 ) . Conclusion p38MAPK specific inhibitor SB203580 improves glomerular filtration rate by reducing oxidative stress.
