摘  要:目的:以白毛夏枯草水提取物、乙酸乙酯提取物为例探究不同提取物的抗肝癌机制。方法:将白毛夏枯草的水提取物(STW)、乙酸乙酯提取物(ZTW)分别作用于肝癌细胞株HepG2和SMMC-7721。运用四甲基偶氮唑盐(MTT)法检测不同浓度的STW、ZTW对肝癌细胞株HepG2和SMMC-7721增生的抑制作用,通过流式细胞仪检测不同浓度的STW、ZTW诱导肝癌HepG2和SMMC-7721细胞凋亡的凋亡率,运用一步法荧光RT-PCR(Real-time PCR)技术检测经不同浓度的STW、ZTW处理后HepG2和SMMC-7721细胞中Bax mRNA的表达情况。结果:STW、ZTW对HepG2和SMMC-7721细胞均有显著的抑制增生和诱导其凋亡的作用,同时能够抑制肝癌血管增殖。STW、ZTW的抑制机制呈一定的浓度依赖性,其中以乙酸乙酯提取物ZTW抑制性最强。结论:白毛夏枯草水提取物、乙酸乙酯提取物均通过抑制肝癌细胞增生,诱导细胞凋亡,抑制肝癌血管增殖,加速肝癌细胞分化等机制来抗肝癌。ObjectiveTo mechanism of anti-hepatoma ajugadecumbens water extractive and ethyl acetate ex-tract as an example to explore the different extracts. Methods:The water extract of Herba etate extract (ZTW) respectively in HepG2 and SMMC-7721 in hepatocellular carcinoma cell yl thiazolyl tetrazolium (MTT) method was used to detect the inhibitory effect of different concentration of STW,ZTW and SMMC-7721 on HepG2 cell proliferation, apoptosis and apoptosis of hepatoma SMMC-7721 cells induced by HepG2 under different concentrations of STW and ZTW were detected by flow cytometry, using one-step fluores-cent RT-PCR (Real-time PCR) expression technique for the detection of the different concentrations of STW, ZTW and HepG2 after treatment of SMMC-7721 cells inBax mRNA. Results : STW andZTW could signiticathe proliteration and induce apoptosis of HepG2 and SMMC-7721 cells, and inhibit the proliteration of hepatoma cells. The inhibitory mechanism of STW and ZTW was concentration dependent, and was the strongest. Conclusion: Herba Ajugae water extract and ethyl acetate extract by inhibiting proliferation ofhepatoma cells, inducecell apoptosis, inhibit vascular proliferation, accelerate cell differentiation mechanisms a-gainst liver cancer.
