漏芦逆转胃癌相关成纤维细胞促癌作用的研究

摘　　要：目的:研究漏芦能否逆转胃癌相关成纤维细胞的促癌作用。方法:漏芦以0.32 g/kg、0.65 g/kg、1.3 g/kg、2.6 g/kg、5.2 g/kg的剂量灌服小鼠,制备含漏芦含药血清。胃癌相关成纤维细胞分别进行常规培养和含漏芦血清培养,制备条件培养液,培养AGS胃癌细胞。用MTT实验、划痕实验和Transwell实验检测AGS细胞增殖、迁移和侵袭能力的变化,用ELISA检测各组胃癌相关成纤维细胞培养液中TGFβ1和IL6含量的变化,以荧光定量PCR检测胃癌相关成纤维细胞中TGFβ1和IL6表达量的变化。结果:常规培养的胃癌相关成纤维细胞显著促进了AGS细胞的增殖、迁移和侵袭;1.3 g/kg、2.6 g/kg和5.2 g/kg组含漏芦血清培养胃癌相关成纤维细胞,使其促AGS细胞增殖、迁移和侵袭的能力明显降低,使其培养液中TGFβ1和IL6的含量明显下降,使其细胞内TGFβ1和IL6 mRNA的表达量明显下调,2.6 g/kg和5.2 g/kg组的效果尤为明显。结论:漏芦能抑制胃癌相关成纤维细胞对TGFβ1和IL6的分泌而逆转其促癌作用。Objective： To investigate whether Rhaponticum uniflorumcan reverse the tumor growth of gastric cancer associated fibroblasts（ GCAF）.Methods： Serums containing drug were prepared from mice after administration of Rhaponticum uniflorum extract（ 1.3 g/kg,2.6g/kg and5.2 g/kg）.Conditioned mediums were collected from normally cultured GCAF and GCAF which were pre-treated with the serums containing Rhaponticum uniflorum.The mediums were used to culture AGS gastric cancer cells.Proliferation,migration and invasion abilities of AGS cells were measured by MTT,scratch and transwell assays,respectively.TGFβ1 and IL6 concentrations of the mediums were assessed by ELISA.TGFβ1 and IL6 mRNA expressions of AGS cells were detected by fluorescence quantitative PCR.Results： The medium from normally cultured GCAF significantly promoted the proliferation,migration and invasion of AGS cells.Rhaponticum uniflorum-contained in serum from the 1.3 g/kg,2.6g/kg and 5.2 g/kg groups decreased the proliferation-,migration-and invasion,reduced the TGFβ1 and IL6 concentrations in GCAF mediums,and down-regulated the TGFβ1 and IL6 mRNA expressions in GCAF cells.And the serum from the 2.6g/kg and5.2 g/kg groups displayed better performance.Conclusion： Rhaponticum uniflorum can reverse the tumor growth of GCAF by inhibiting its secretion of TGFβ1 and IL6,and is valuable for gastric cancer therapy.
