定心胶囊通过STAT3通路改善Ang Ⅱ诱导的ACE2-/y小鼠心室舒缩障碍的机制研究

摘　　要：目的:研究定心胶囊对ACE2-/y小鼠血压升高和心功能异常的作用及其调控机制。方法:应用血管紧张素Ⅱ(Ang Ⅱ)静脉注射ACE2-/y小鼠建立高血压及心功能障碍模型,实验小鼠随机分为ACE2+/y对照组、ACE2-/y模型组、定心胶囊组(12.5g/kg)和卡托普利组(3.75mg/kg)。造模后,监测小鼠收缩压,检测心功能,观察信号转导及转录活化因子3(STAT3)和STAT1的表达及活化。结果:定心胶囊降低血浆Ang Ⅱ水平,显示持续的降压效果。心脏超声多普勒显示显著增加短轴缩短率和应变率、降低LVEDd和LVSDd并改善左室前壁厚度。蛋白免疫印迹显示定心胶囊降低STAT3的磷酸化并抑制其核转位。结论:定心胶囊改善Ang Ⅱ诱导的血压升高及心功能障碍,与其抑制STAT3的磷酸化及核转位有关。Objective： To explore the effects of Dingxin Capsule on the hypertension and cardiac dysfunction in mice induced by angiotensin Ⅱ（ Ang Ⅱ）.Methods： ACE2-/ymice were infused with Ang Ⅱ to establish hypertension and ventricular dysfunction model.Animals were divided into ACE2＋/y（Control）,ACE2-/y（ Model）,Dingxin Capsule（ 12.5g/kg） and Captoril（ 3.75mg/kg） groups.The systolic pressure and cardiac function were measured and the activation of signal transducers and activators of transcription 3（ STAT3） were detected.Results：Dingxin capsule lowered the plasma Ang Ⅱ level and systolic pressure.It increased fractional shortening and velocity of circumferential fibre shortening and decreased left ventricular end-diastolic diamension and LV end-systolic diamension.Dingxin capsule ameliorated anterior left ventricular wall thickness.It reduced the phosphorylation of STAT3 and inhibited its translocation to neclear.Conclusion： Dingxin capsule ameliorated the hypertension and ventricular dysfunction induced by Ang Ⅱ which may partly attribute to the inhibition of phosphorylation and translocation of STAT3.
