摘  要:目的:探讨天芪益智颗粒对痴呆模型大鼠认知功能及抗氧化能力的影响。方法:建立鹅膏蕈氨酸基底核微量注射AD模型。采用Morris水迷宫法检测大鼠的学习记忆能力变化;尼氏染色法检测海马和大脑皮层神经元病理改变;分光度法检测大鼠脑组织MDA、H_2O_2含量及SOD、GR、CAT活力。结果:与模型组相比,天芪益智颗粒0.63g/kg、0.42g/kg剂量组第5d逃避潜伏期明显降低,0.42g/kg剂量组穿越平台次数明显增加,神经元数量增多;与模型组比较,天芪益智颗粒0.63g/kg、0.42g/kg剂量组显著降低MAD、H_2O_2含量及提高GR活力。结论:天芪益智颗粒具有改善模型大鼠认知功能障碍,保护神经细胞的作用,该作用与其增强抗氧化能力有关。Objective: To investigate effects of Tianqiyizhi Granules on cognitive function and antioxidant ability in AD rats.Methods: SD rats were micro-injected with Ibotenic acid into their basal ganglia to establish AD models.Learning and memory ability were tested by the Morris water maze test; neurons in the hippocampus and cerebral cortex were analyzed by nissl staining; The levels of MAD,H2O2 and activities of SOD,GR,CAT in rat brain tissue were detected with spectrophotometry.Results: Compared with the model group,Tianqiyizhi Granules at 0.63g/kg,0.42g/kg dose could significantly reduce the escape latency on 5th day,Tianqiyizhi Granules at 0.42g/kg dose could significantly increase the crossing platform frequency and the number of neurons rose.Compared with the model group,Tianqiyizhi Granules at 0.63g/kg,0.42g/kg dose could significantly reduce the levels of MAD,H2O2 and significantly improve the activity of GR,and also could slightly increase the activity of SOD,CAT.Concusion: Tianqiyizhi Granules could ameliorate cognitive impairment and protect neurons through enhancing antioxidant ability.
