益智仁-乌药醚提取物对糖尿病肾病小鼠p27kip1蛋白表达的影响

摘　　要：目的:观察益智仁-乌药醚提取物对糖尿病肾病小鼠p27kip1蛋白表达的影响。方法:应用STZ造模,建立糖尿病肾病模型小鼠,分别给予厄贝沙坦10mg/(kg·d)、益智仁-乌药醚提取物3个剂量,即20g/(kg·d)、10g/(kg·d)、5g/(kg·d)治疗4周,采用RT-PCR和Western blotting方法观察各组小鼠p27kip1蛋白的表达情况。结果:与正常对照组比较,糖尿病肾病模型组p27kip1蛋白表达明显上调;与模型组比较,益智仁-乌药醚提取物20g/kg组和厄贝沙坦组p27kip1蛋白表达均明显下调。结论:益智仁-乌药醚提取物对糖尿病肾病小鼠肾脏有一定的保护作用,其机制可能与调节p27kip1蛋白表达有关。Objective： To observe the influence of ether extract of the Yizhiren-Wuyao on p27kip1 Protein Expression in diabetic nephropathy mice. Methods ： STZ was applied to establish the model of diabetic nephropathy mice. Diabetic nephropathy mice were given positive control drug irbesartan 10mg/（ kg · d） ,ether extract of the Yizhiren-Wuyao three intervendonal 20 g/（ kg ·d） , 10 g/（ kg · d） ,5g/（ kg · d）for 4 weeks. The expression of p27kip1 Protein was observed by RT-PCR and Western blotting method. Results： Compared with the normal control group, diabetic nephropathy increased expression of p27kip1 Protein in the model group （P 〈 0.01 ） ; compared with the model group,in the ether extract of the Yizhiren-Wuyao 20g/kg group and irbesartan group p27kip1 Protein expressions significantly decreased （ P 〈 0.01 ）. Conclusion ： ether extract of the Yizhiren-Wuyao on kidney of diabetic nephropathy mice has a protective effect, its mechanism may be related to reg- ulating the expression of p271dpl protein.
