摘 要:目的:探讨复方皂矾洗剂对雄激素性脱发的作用及机制。方法:60只雄性成年SD大鼠随机均分为空白组,模型组,米诺地尔酊0.05g/ml组,复方皂矾洗剂2g/ml组,复方皂矾洗剂0.5g/ml组,复方皂矾洗剂0.25g/ml组6组中,颈后皮下注射丙酸睾酮制造雄激素脱发大鼠模型。造模时予以背部外用药物,空白组、模型组均予以蒸馏水,余下4组分别予以0.05g/ml的米诺地尔酊,2g/ml、0.5g/ml、0.25g/ml的复方皂矾洗剂。计数皮肤横切面毛囊数量;测定皮肤中甘油三酯、游离脂肪酸、总胆固醇含量,评价不同浓度复方皂矾洗剂疗效。测定血液中睾酮、二氢睾酮的含量,测定皮肤中二氢睾酮的含量以及固醇调节元件结合蛋白(SREBP-1、SREBP-2)及含半胱氨酸的天冬氨酸蛋白水解酶-12(caspase-12)的表达,探讨复方皂矾洗剂作用机制。结果:复方皂矾洗剂2g/ml组中总毛囊和终毛毛囊数量较模型组增多,甘油三脂、游离脂肪酸、总胆固醇的含量较模型组降低,复方皂矾洗剂0.5g/ml组,复方皂矾洗剂0.25g/ml组作用程度依次减弱。2g/ml复方皂矾洗剂组皮肤中二氢睾酮的含量,SREBP-1、SREBP-2及caspase-12的表达均较模型组降低。结论:2g/ml、0.5g/ml、0.25g/ml的复方皂矾洗剂对雄激素性脱发均有治疗作用,以2g/ml为最佳剂量。复方皂矾洗剂的作用机制可能与降低皮肤中二氢睾酮的含量和SREBP-1,SREBP-2的表达,降低油脂分泌,抗脂质过氧化,抑制内质网应激,抑制毛囊内质网途径的凋亡有关。Objective: To investigate the effect and mechanism of the Compound Zaofan Lotion on androgenetie alopecia. Methods: Sixty healthy male adult SD rats were randomly equally divided into control group, model group, 0.05g/ml Minoxidil Tincture group, 2g/ml Compound Zaofan Lotion group,0.5g/ml Compound Zaofan Lotion group, 0.25g/ml Compound Zaofan Lotion group. Subcutaneous injection of testoster- one propionate was used to establish the model of androgenetic alopecia. The control group and model group were given distilled water in the skin of the back, and the remaining four groups were given 0.05g/ml Minoxidil Tincture, 2g/m1,0.5g/m1,0.25g/ml Compound Zaofan Lo- tion. The number of cross sections of hair follicle in the skin was counted, the content of triglyceride, free fatty acid and total cholesterol were determined to evaluate the therapeutic effect of Compound Zaofan Lotion. The content of testosterone, two hydrogen testosterone in the blood, two hydrogen testosterone and expressions of SREBP-1, SREBP-2 and caspase-12 were detected in the skin to investigate the mechanism of Compound Zaofan Lotion on androgenetic alopecia rats. Results: The number of the total hair follicles and the terminal hair follicles in 2g/ml Compound Zaofan Lotion group was higher than that in the model group,the contents of triglycefide, free fatty acid and total cholesterol in 2g/ ml Compound Zaofan Lotion group were lower than those in the model group, they slightly decreased in 0.5g/ml Compound Zaofan Lotion group and 0.25g/ml Compound Zaofan Lotion group. The content of two hydrogen testosterone, the expression of SREBP-1, SREBP-2 and caspase-12 in the Compound Zaofan Lotion group were lower than those in the model group. Conclusion: 2g/m1,0.5g/ml,0.25g/ml Com- pound Zaofan Lotion have therapeutic effect on androgenetic alopecia,2g/ml is the optimal dose. The mechanism of Compound Zaofan Lotion may be that it can reduce the content of two hydrogen testosterone and the expression of SREBP-I and SREBP-2 in skin, reduce the oil
