摘  要:目的:观察肾复宁颗粒对慢性肾功能衰竭大鼠的作用及可能机制。方法:SD大鼠采用右肾切除左肾外动脉分支结扎方法建立CRF模型,随机分为模型对照组、雷公藤多甙片组(0.006g/kg)、肾复宁颗粒组(9g/kg)、肾复宁颗粒组(6g/kg)、肾复宁颗粒组(3g/kg),各15只,另设假手术组15只,连续给药8周,观察血清Scr、BUN、Cys-C含量及肾脏组织中TGF-β1mRNA、细胞凋亡基因Fas/Fas L表达水平。结果:治疗后,治疗组及雷公藤多甙片组大鼠血清中Scr、BUN、Cys-C水平明显下降,大鼠肾脏组织中TGF-β1 mRNA、Fas/Fas L表达减少,尤以肾复宁颗粒(9g/kg)显著。结论:肾复宁颗粒通过抑制慢性肾功能衰竭大鼠肾脏TGF-β1mRNA、Fas/Fas L表达,调控肾实质细胞调亡,从而改善肾功能,达到延缓肾脏纤维化作用,其疗效具有一定剂量依赖性。Objective: To observe the effect and possible mechanism of Shenfuning Granules on chronic renal failure (CRF)in rats. Methods: CRF rat model was established by the Right nephrectomy and left renal artery ligation, and they were randomly divided into the model control group (SD), sham operated group, the glycosides group (0. 006g/kg), Shenfuning Granules group (9g/kg), Shenfuning Granules group (6g/kg), and Shenfuning Granules group (3g/kg), 15 rats in each group. The treatment lasted for 8 weeks. Contents of Scr, BUN and Cys- C in serum and levels of TGF-β1mRNA,apoptosis gene Fas/FasL were measured in renal tissues. Results: After the treatment, the serum levels of Sc, BUN, Cys-C decreased in the treatment and glycosides tablets group, compared with the model group, differences were signifi- cant. Expressions of TGF-β1 mRNA and Fas/FasL significantly decreased in the kidney of rats, especially in the dose of Shenfuning Granules (9g/kg). Conclusion: Shenfuning Granules can inhibit expressions of TGF-β1 mRNA and Fas/FasL in the kidney of the CRF rats, regulate the apoptosis of renal parenchymal cells, improve renal function and delay the renal fibrosis. The efficacy has a certain dose dependence.
