摘  要:目的探讨曼陀罗子对小鼠外周血、骨髓嗜多染红细胞微核率和精子畸变的影响。方法将昆明种小鼠按体重随机分为阴性对照组、阳性对照组、曼陀罗子药粉组、曼陀罗子总碱组。阴性对照组小鼠以0.02 mL·g^(-1)·d^(-1)的0.5%CMC-Na灌胃,阳性对照组以0.02 g·kg^(-1)·d^(-1)的环磷酰胺(CP)灌胃,曼陀罗子药粉组分别以2.5、5和10 g·kg^(-1)·d^(-1)的曼陀罗子药粉灌胃,曼陀罗子总碱组以1.25、2.5和5 g·kg^(-1)·d^(-1)的曼陀罗子总碱提取物灌胃,连续给药14 d,每隔2 d称重。Giemsa染色法观察小鼠外周血和骨髓嗜多染红细胞微核率,伊红染色法观察小鼠精子畸变率。结果给药组小鼠的体重增量、睾丸和附睾指数均较对照组显著降低,而骨髓和外周血微核率及精子畸变率显著增加。结论10 g·kg^(-1)·d^(-1)的曼陀罗子药粉和1.25、2.5、5 g·kg^(-1)·d^(-1)的曼陀罗子总碱提取物对小鼠可能具有遗传毒性。Objective To investigate the influence of daturae semen on micronuclei formation in bone marrow and peripheral blood polychromatic erythrocyte. Methods Kunming strain mice were randomly divided into negative control group, positive control group, semen daturae powder group and total alkaloids of daturae semen group. The negative control group mice were treated with 0.5% CMC-Na and positive control group with 0.02 mL ·kg^-1·d^-1 CP.daturae Semen powder group were administered intragastrically by 2.5, 5 and 10 g·kg^-1·d^-1 of daturae semen powder, and total alkaloids of daturae semen group by 1.25-2.5 and 5 g·kg^-1·d^-1 of total alkaloids of daturae semen . Mice body weight were measured every three days, and micronucleus in bone marrow and peripheral blood were analyzed by Giemsa staining and sperm abnormality by Eosin staining. Results Compared with negative control group, daturae semen could significantly decrease the weight increments of mice, and increase the micronucleus in bone marrow and peripheral blood and sperm abnormality in mice. Conclusion 10 g·kg^-1·d^-1 of daturae semen powder and 1.25, 2.5, 5 g·kg^-1·d^-1 of total alkaloids of semen daturae may have genetoxicity to mice.
