摘 要:为了提高玉米愈伤组织再生率,以1个玉米自交系和3个杂交种的幼胚愈伤组织为试材,接入含有不同浓度2,4-D的分化预处理培养基(N6+脯氨酸700 mg/L+水解酪蛋白500 mg/L+蔗糖30 g/L),2,4-D浓度设0、0.5、1.0和1.5 mg/L计4个处理,培养7 d后转移到分化培养基中进行分化处理,以继代培养7 d的愈伤组织直接转入分化培养基进行植株再生作为对照,研究了不同浓度2,4-D分化预处理对玉米愈伤组织再生率和再生进程的影响。结果表明:愈伤组织在转入分化培养基前进行预处理,可以提高玉米植株再生率。分化预处理培养基以不加2,4-D的效果最好,玉米愈伤组织再生率最高,与对照差异达到了极显著水平,且分化成苗进程加快。不同基因型中,杂交种87-1×郑22的植株再生率最高,与其他2个杂交种差异不显著,但三者的植株再生率均极显著高于自交系87-1。对玉米愈伤组织进行分化前的预处理,可以提高植株再生率,加快再生进程。In order to improve the regeneration rate on maize callus, different 2,4-D concentration including 0, 0.5, 1.0 and 1.5 mg/L were used to pretreatment callus media of N6+700 mg/L proline+500 mg/L casein hydrolysate+30 g/L sucrose. The regenerate rate of different media after 7 days transformation was compared with CK as directly transformation. The effects of different 2,4-D concentration of differentiation pretreatments on callus regeneration frequency and regeneration process were investigated by using one inbred line and three hybrids immature embryos of maize as experimental materials. The results indicated that there was a significant increase on regeneration frequency with different 2,4-D concentration of pretreatments before transferred to differentiation medium The pretreatment medium without 2,4-D could significantly improve the maize regeneration seedling frequency and accelerate the plant regeneration process. There was a significant different regeneration frequency with various genotype calluses. With the increase of 2,4-D concentration, the regeneration frequency of the same genotype callus was in a downward tendency in the several experimental genotypes. There was a highest regeneration rate on hybrids 87-1×Zheng 22 and no significant difference with other 2 hybrids. The plant regeneration frequency of three hybrids was generally higher than that of inbred line of 87-1. This demonstrated that the pretreatment with maize callus can increase the plant regeneration frequency and accelerate the regenerate process.
