摘 要:本研究采用响应面方法优化重组人对氧磷酶1(Recombinant Human Paraoxonase 1,rhPON1)在毕赤酵母中的表达。首先通过研究甲醇浓度、p H、培养基装液量3个单因素对rhPON1表达的影响,然后以rhPON1单位体积酶活力为响应值进行响应面分析。结果显示:甲醇浓度1.42%,p H值5.91,培养基装液量9.39%是最优发酵条件。在此优化条件下,rhPON1的单位体积酶活力预测值为0.737U/mL,与验证值(0.735U/mL)基本一致。人对氧磷酶1可以水解多种有机磷化合物,被认为是一种有机磷农药中毒的解毒剂。In this study, the expression of Recombinant Human Paraoxonase 1 (rhPON1) in Pichia pastoris was optimized by response surface method. A single-factor test coupled with response surface methodology (RSM) was used to adjust the fermentation conditions. The results showed that the optimized induction conditions were as fol- lows: methanol concentration of 1.42%, pH 5.91, culture volume of 9.39%, and the predicted theoretical enzyme activity of rhPON1 was 0.737U/mL. Results of the verification experiment under the optimized fermentation condi- tions showed that the enzyme activity of rhPON1 was 0.735U/mL. Human paraoxonase 1 (hPON1) is considered as an potential organophosphate pesticide scavenger which can hydrolyze various organophosphates.
