摘 要:目的:探讨Notch信号通路配体Jagged1、Dll4在子痫前期胎盘组织中的表达及意义。方法:选取轻度子痫前期患者30例(轻度组)、重度子痫前期患者30例(重度组)、正常妊娠孕妇30例(对照组),采用免疫组化法检测其胎盘组织中Jagged1、Dll4的定位表达,采用荧光定量PCR技术和Western blot法分别检测胎盘组织中Jagged1、Dll4 mRNA及蛋白的表达。结果:Jagged1主要表达于血管内皮细胞和绒毛滋养层细胞,Dll4主要表达于血管内皮细胞。轻、重度组胎盘组织中Jagged1、Dll4 mRNA及蛋白的表达水平均低于对照组(P<0.05),且重度组低于轻度组(P<0.05);对照组胎盘组织中Jagged1与Dll4在mRNA和蛋白水平上的表达均呈正相关(r=0.806和0.808,P<0.05),轻度组中两者表达的相关性减弱(r=0.675和0.560,P<0.05),而在重度组中未发现两者的相关性。结论:Jagged1、Dll4在子痫前期胎盘组织中表达失衡,与子痫前期的发病关系密切。Aim: To explore the expressions of Notch ligand Jagged1 and Dll4 in preeclampsia placentas. Methods:Placental specimens from 30 normal pregnant women( control group),30 mild preeclampsia women( mild group) and 30 severe preeclampsia women( severe group) were collected to detect the location of Jagged1 and Dll4 protein by immunohisto chemistry; RT-qPCR and Western blot were used to determine the expressions of mRNA and protein of Jagged1 and Dll4,respectively. Results: Jagged1 was mainly expressed in vascular endothelial cells and trophoblast cells. Dll4 was expressed in vascular endothelial cells. The mRNA and protein expressions of Jagged1 and Dll4 in placentas from patients of mild group and severe group were significantly lower than those from control group( P 〈 0. 05),which was lowest in severe group( P 〈 0. 05). The mRNA and protein expressions of Jagged1 were positively correlated with those of Dll4 in control group( r = 0. 806,0. 808,P 〈 0. 05),and weakened in mild group( r = 0. 675,0. 560,P 〈 0. 05),but had no correlation in severe group. Conclusion: The imbalance of Jagged1 and Dll4 may be related to the occurrence of preeclampsia.
