摘 要:目的:观察中药复方健脾清化方对糖尿病模型大鼠血糖波动幅度和糖脂代谢的影响。方法:实验分为正常组、模型组和健脾清化方组,除正常组外均采用小剂量链脲佐菌素联合高脂高糖饲料诱导建立Wistar大鼠糖尿病模型。健脾清化方组以13 g·kg-1·d-1的剂量灌胃,正常组和模型组分别给予同体积的蒸馏水灌胃,共干预4周。采用葡萄糖氧化酶方法检测大鼠血清葡萄糖,快速血糖仪测定大鼠尾尖血糖;通过公式计算血糖波动相关指数,包括平均血糖波动幅度(mean amplitude of glycemic excursions,MAGE)、最大血糖波动幅度(largest amplitude of glycemic excursions,LAGE)及M值;腹腔糖耐量检测糖代谢,包括腹腔糖耐量(IPGTT)、曲线下面积(AUCg)、平均血糖;ELISA法测定脂代谢,包括总胆固醇(TC)、低密度脂蛋白(LDL)、高密度脂蛋白(HDL)、三酰甘油(TG)。结果:与模型组比较,健脾清化方能明显降低糖尿病大鼠IPGTT各时间点血糖、平均血糖和AUCg(P<0.05,P<0.01);健脾清化方能有效改善糖尿病大鼠血糖波动,降低MAGE、M值(P<0.05,P<0.01);健脾清化方能有效改善脂代谢,降低糖尿病大鼠的TG水平(P<0.05)。结论:健脾清化方能有效改善糖尿病模型大鼠的血糖波动,并对糖尿病模型大鼠的糖脂代谢也具有一定的改善作用。Objective : To observe the influence of Jianpi Qinghua Recipe on blood glucose fluctuation and metabolism of glucose and lipid in diabetic rats. Methods : Diabetes model was induced in Wistar rats by streptozotocin at low dose combined with high fat and high sugar diet. The rats were divided into the normal group, model group and Jianpi Qinghua Recipe(JPQH) group. The JPQH group was treated with Jianpi Qinghua Recipe at daily dose of 13 g/kg for intragastric administration, the other groups were treated with distilled water at the same volume, with a course of g weeks. The glucose level in rat serum was detected by glucose oxidase method, and glucose level in rat tail tip was detected by fast blood glucose meter. The related indexes of blood glucose fluctuation were calculated by formula, including mean amplitude of glycemic excursions ( MAGE ), largest amplitude of glycemic excursions(LAGE) and M value. The glucose metabolism was evaluated by intraperitoneal glucose tolerance test (IPGTY), including intraperitoneal glucose tolerance, area under curve (AUGg), mean blood glucose. The lipid metabolism was evaluated by ELISA, including total cholesterol (TC) ,low density lipoprotein (LDL), high density lipoprotein (HDL) ,triacylglycerol (TG). Results: Compared with the model group, Jianpi Qinghua Recipe could significantly decrease the level of blood glucose ,mean blood glucose and AUCg in diabetic rats at each timepoints in IPGTT( P 〈 0.05 ,P 〈 0.01 ). Jianpi Qinghua Recipe could effectively improve the blood glucose fluctuation in diabetic rats, decrease MAGE and M value ( P 〈 0.05, P 〈 0.01 ). Jianpi Qinghua Recipe could effectively improve the lipid metabolism, decrease the TG level in diabetic rats(P 〈0.05). Conclusion: Jianpi Qinghua Recipe can effectively improve the blond glucose fluctuation in diabetic rats,and shows a certain effects on improving the metabolism of glucose and lipid.
