摘 要:目的通过建立慢性心衰大鼠模型,观察养心汤对慢性心力衰竭模型大鼠血浆肾素活性(PRA)、血管紧张素Ⅱ(AngⅡ)、醛固酮(ALD)的影响。方法 SD雄性大鼠70只,随机选取10只大鼠作为空白对照组,其余60只大鼠腹腔注射阿霉素每周4 mg/kg,连续6周,6周后全部大鼠予心脏超声测定心功能,以LVEF<60%为心力衰竭模型复制成功;将模型复制成功大鼠按随机数字表法分为模型对照组、养心汤组、卡托普利组。空白对照组、模型对照组予等体积的蒸馏水灌胃;养心汤组予养心汤浸膏;卡托普利组予卡托普利,连续4周,实验结束后,大鼠腹主动脉采血测定血清PRA、AngⅡ、ALD水平及心肌HE染色观察心肌组织病理形态学改变。结果与空白对照组比较,模型对照组大鼠血清PRA、AngⅡ、ALD水平增加(P<0.01);与模型对照组比较,养心汤组、卡托普利组大鼠血清PRA、AngⅡ、ALD水平降低(P<0.01);与卡托普利组比较,养心汤组大鼠血清PRA、AngⅡ、ALD水平升高,但差异无统计学意义(P>0.05)。结论养心汤能降低慢性心力衰竭模型大鼠血清PRA、AngⅡ、ALD水平,抑制RAAS活性,保护心肌组织,延缓心室重构,其疗效与卡托普利相当。Objective: To observe the effect of Yangxin Decoction on PRA,AngⅡ and ALD by establishing rat model of chronic heart failure. Methods:10 rats were selected as the blank control group among 70 ones,and the remaining 60 ones received intraperitoneal injection of adriamycin 4 mg/kg per week,for 6 weeks. 6 weeks later, all rats received cardiac ultrasound measurement of cardiac function,with LVEF〈60% for heart failure model es-tablished successfully. The successful models were randomly divided into the model control group,Y angxin De-coction group,and Captopril group. The blank control group and the model control group were given equal volume of distilled water by gavage;Yangxin Decoction group was given Yangxin Decoction extract;Captopril group was given Captopril,for 4 weeks. After the experiment,blood samples were obtained from abdominal aorta to detect PRA,AngⅡ and ALD level and morphological changes of myocardial tissue were observed by myocardial HE staining. Results: Yangxin Decoction had an effect on serum PRA,AngⅡ and ALD level of rats with chronic heart failure. Compared with the blank control group,the levels of serum PRA,AngⅡand ALD increased in mod-el control group (P〈0.01);compared with the model control group,the levels of serum PRA,AngⅡ and ALD in Yangxin Decoction group and Captopril group were decreased(P〈0.01);compared with Captopril group,the levels of serum PRA,AngⅡ and ALD increased in Yangxin Decoction group,but the difference was not statistically sig-nificant (P〉0.05). Conclusion: Yangxin Decoction can reduce serum PRA,AngⅡ and ALD level of rats with chronic heart failure,inhibit the activity of RAAS,protect myocardial tissue,and delay ventricular remodeling. The curative effect is quite the same as Captopril.
