弓形虫急性感染对小鼠学习能力的影响及其机制

摘  要:目的探索处于弓形虫急性感染期的雌、雄小鼠的学习记忆能力的变化及其可能的机制。方法以弓形虫RH株速殖子数量300/只腹腔注射小鼠建立弓形虫急性感染期模型,利用穿梭箱实验对感染组小鼠和对照组小鼠感染后第0、1、2、3、4和5天的学习记忆能力进行评价,并从感染当天开始,每天分离各组小鼠中脑,以荧光定量PCR法对其中脑内酪氨酸羟化酶(TH)的表达水平进行检测。结果从感染后第3天开始,感染组小鼠的学习记忆能力明显低于对照组小鼠,差异有统计学意义(第3、4和5天:P<0.05)。从感染后第1天开始,感染组小鼠中脑TH表达量开始升高,感染组平均表达量相较于对照组明显升高,差异有统计学意义(P<0.05),且雌性感染组TH平均表达量高于雄性感染组,差异有统计学意义(P<0.05),而对照组之间则无明显区别(P>0.05)。结论弓形虫急性感染可损害小鼠的学习记忆能力并使其中脑TH表达水平增高,该作用具有性别差异且雌性更易受影响。Objective To detect the changes on learning ability of mice when suffered with Toxoplasma gondii during acute phase with regard to both genders, and explore the possible mechanism underlying these changes. Methods The mice acute infection model was established by injecting tachyzoites intraperitoneally at 300 per mouse ; the learning ability of both male and female mice were evaluated every day in the time of experiment period. Themice midbrains were collected for the study. The relative transcription levels of tyrosine hydroxylase (TH) were detected by using fluorescent real- time PCR. Results The learning ability of infected mice was significantly lower than that of control mice (at the 3^rd, 4^th and 5^th day, P〈O.05 ). From the 1^st day post-infection, the levels of mRNA of TH in mice midbrain were increased. The average level of TH mRNA in infection group was significantly higher than that in control group (P〈0.05). Female gained higher level of TH mRNA than male (P〈0.05). And there was no significant differences between male and female of control group (P〉0.05). Conclusion Toxoplasma gondii infection could impair the learning ability as well as the mRNA level of TH in midbrain, and show a sex-specific results that females seem more susceptible.
