摘 要:本研究将齐墩果酸(OA)与5-氟尿嘧啶(5-Fu)联合作用于A549细胞,用HPLC法测定有无齐墩果酸作用后细胞内外5-Fu的浓度,观察药物浓度的变化情况。结果显示当未给予OA时,刚开始5-Fu浓度与作用时间呈正相关,但增加到一定浓度的时候,其细胞内的浓度基本没有显著的变化,由此可能出现耐药性的情况。当给予OA后,细胞内的药物浓度一开始时就出现比较高的浓度,而随着作用时间的增长,细胞内的浓度并没有增加,呈现一个相对比较稳定的范围,由此推测可能是OA影响了细胞膜对5-Fu的转运。由此推测齐墩果酸可能会延长5-氟尿嘧啶的耐药性产生时间,并能与5-氟尿嘧啶具有协同的的抗癌细胞效果。In this study, oleanolic acid (OA) combined with 5- fluorouracil (5-Fu) was applied to A549 cells, and the concentration of intra and extra cellular 5-Fu after the effect of OA was investigated by HPLC, and we observed the change of drug concentration. The results showed that when not given OA, the concentration of 5-Fu and the reaction time were positively correlated at the beginning, but when the concentration of 5-Fu increased to a certain level, it basically had no significant changes in cells, which might appear drug resistance. When given OA, the drug concentration within the cell had relatively high levels at first, but with the increase of time, the intracellular concentration did not increase, showing a relatively stable range, from which we predicted that OA might affect the transport of 5-Fu on the cell membrane. Therefore, we speculated that OA might prolong the drug resistance of 5-Fu and produce synergistic anti-cancer cell effects with 5-Fu.
