摘 要:目的:观察中药复方丹瓜方对高脂糖尿病大鼠离体心脏的垂体后叶素所致心肌缺血模型的影响,并探讨其可能的作用机制。方法:选取SD大鼠,随机分出正常组大鼠,其余大鼠经糖尿病造模后,随机分为模型组、糖脉康组及丹瓜方低、中、高剂量组。干预12周后采用Langendorff恒流灌流装置及加有垂体后叶素的K-H液灌注离体心脏,分别观察并记录各时间段的冠脉流量、心率及心肌收缩幅度。结果:与正常组相比,中、高剂量组第30分钟心肌收缩幅度均升高(P<0.05);与高剂量组相比,低剂量组和模型组第10分钟心率均降低(P<0.05),低剂量组、模型组及正常组第30分钟心率均降低(P<0.05);与中剂量相比,正常组及模型组第15分钟心肌收缩幅度均下降(P<0.05或P<0.01)。结论:丹瓜方对糖尿病缺血缺氧心肌有一定的保护作用。Objective : By observing influence of Chinese medicine compound Dangua Recipe on the isolated heart of mice with diabetes mellitus fed by high fat diet underwent myocardial ischemia caused by pituitrin to study the possible mode of action. Methods : The rats of normal group were separated randomly from all the selected SD rats. The rest of the rats were divided into the model group, the Tangmaikang group, the low dose group of Dangua Recipe, the medium dose group of Dangua Recipe and the high dose group of Dangua Recipe. After diabetes mellitus rats were induced successfully, each group was intragastrically adminis- trated for 12 weeks. Thereafter, the isolated heart of rats were disposed by using the langendorff apparatus to pour the KH liquid with pituitrin into the hearts. Simultaneously observe and record respectively the coronary flow, the heart rate and the degree of myocardial contraction in each period. Results : Compared with the normal group, the degree of myocardial contraction of the high dose group and the medium dose group at the 30 -minute mark was bigger(P 〈 0. 05 ). Compared with the high dose group, the heart rate of the low dose group at the 10 - minute mark was lower( P 〈 0. 05 ). The heart rates of the low dose group, the medium dose group and the normal group at the 30 - minute mark were lower(P 〈 0. 05 ). Compared with the medium dose group, the de- gree of myocardial contraction of the normal and the model group were smaller(P 〈0. 05 or P 〈0. O1 ). Conclusion:Dangua Reci- pe had some protective effect on the anoxic cardiac muscle of the diabetic rats.
