小肠型脂肪酸结合蛋白基因G54A单核苷酸多态性对肥胖患者脂代谢的影响

摘  要:目的 探讨小肠型脂肪酸结合蛋白(FABP2)基因G54A的单核苷酸多态性与肥胖的关系,分析突变型54A FABP2基因对脂代谢的影响.方法 筛选84例肥胖患者和正常对照组60名,采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)方法对FABP2基因G54A基因型进行检测,并分析等位基因分布频率.用酶法检测空腹血浆TG、HDL-C和LDL-C浓度.结果 FABP2基因G54A多态性频率分布为:肥胖组G/G、G/A、A/A基因频率分别为19.0%(16/84)、73.8%(62/84)、7.2%(6/84),正常对照组G/G、G/A、A/A基因频率分别为38.3%(23/60)、58.3%(35/60)、3.3%(2/60),两组基因频率比较,差异有统计学意义(χ2=6.97,P〈0.05).肥胖组54G、54A等位基因频率分别为56.0%(94/168)和44.0%(74/168),正常对照组54G、54A等位基因频率分别为67.5%(81/120)和32.5%(39/120),两组等位基因分布频率差异有统计学意义(χ2=3.92,P〈0.05).肥胖组A/A突变纯合子型携带者血浆TG(2.36±0.82) mmol/L、LDL-C(3.94±0.96) mmol/L和G/A 突变杂合子型携带者血浆TG(2.06±0.59) mmol/L,LDL-C(3.29±0.55) mmol/L浓度均显著高于G/G野生型携带者血浆TG(1.61±0.67) mmol/L、LDL-C(2.86±0.75) mmol/L,差异均有统计学意义(t值分别为2.206、2.575、2.632、2.278,P均〈0.05),而血浆HDL-C浓度A/A突变纯合子型携带组(1.23±0.34) mmol/L、G/A 突变杂合子型携带组(1.21±0.26) mmol/L与G/G野生型携带者(1.29±0.31) mmol/L比较差异均无统计学意义(P均〉0.05).肥胖组中,与G/A 突变杂合子型携带者血浆TG(2.06±0.59) mmol/L、HDL-C(1.21±0.26) mmol/L、LDL-C(3.29±0.55) mmol/L浓度比较,A/A突变纯合子型携带者血浆LDL-C(3.94±0.96) mmol/L浓度升高,差异有统计学意义(t=2.476,P〈0.05),而TG(2.36±0.82) mmol/L和HDL-C(1.23±0.34) mmol/L两组间差异均Objective To explore the relationship between intestinal fatty acid binding protein(FABP2) gene G54A polymorphism and obesity,the effect of mutant 54A FABP2 gene on serum lipids metabolism.Methods The total of 84 subjects with obesity and 60 subjects with normal weight were involved in this study.The G54A FABP2 gene allele and genotype frequencies were detected by polymerase chain reaction (PCR)-restriction fragment length polymorphism(RFLP) technology.The automatic biochemical Analyzer was used to detect triglyceride(TG),high-density lipoprotein cholesterol(HDL-C) and low-density lipoprotein cholesterol(LDL-C) levels.Results The results of study on FABP2 gene polymorphism revealed as followed:in obese groups,the frequencies of GG,GA,A/A genotypes was 19.0%(16/84),73.8%(62/84) and 7.2%(6/84),respectively;in control group,the frequencies of G/G,G/A,A/A genotypes was 38.3%(23/60),58.3%(35/60),3.3%(2/60),respectively;the differences between two groups was statistically significant(χ2=6.97,P〈0.05).In obesity group,the frequencies of alleles were 56.0%(94/168) for 54G and 44.0%(74/168) for 54A;in the control group,the frequencies of alleles were 67.5%(81/120) for 54G and 32.5%(39/120) for 54A;the differences between two groups was statistically significant(χ2=3.92,P〈0.05).The plasma biochemical variables results showed as followed:in obesity group,the carriers of A/A homozygous genotypes and G/A heterozyous genotypes both had significantly higher plasma TG((2.36±0.82) mmol/L,(2.06±0.59) mmol/L,respectively)and LDL-C((3.94±0.96) mmol/L,(3.29±0.55) mmol/L,respectively)level than those with GG wild genotype(t=2.206,2.575,2.632,2.278;P〈0.05).The level of HDL-C in the carriers of A/A homozygous genotypes((1.23±0.34) mmol/L)and in the carriers of G/A heterozyous genotypes((1.21±0.26) mmol/L) were not significantly different than those with GG wild genotype�
