摘 要:采用光谱法研究了金属离子Cu^(2+)、Zn^(2+)和Fe^(3 +)对盐酸二氧丙嗪(Dioxopromethazine hydrochloride,DPZ)与人血清白蛋白(Human serum albumin,HSA)相互作用的影响.结果表明,在Cu^(2+)、Zn^(2+)和Fe^(3 +)等离子存在条件下,DPZ与HSA相互作用的结合常数、结合位点数及结合距离数值均有不同程度的增加.结合常数的提高表明金属离子的存在能提高DPZ与HSA的结合能力,导致DPZ的最大作用强度降低,使药物的作用时间延长.HSA的构象没有因为与Cu^(2+)、Zn^(2+)和Fe^(3 +)的结合而改变,表明DPZ与HSA结合能力的增加是因为形成的DPZ-HSA-金属离子三元复合物比DPZ-HSA二元复合物更为稳定.The effects of Cu^2+ , Zn^2+ and Fe^2+ on the interaction between dioxopromethazine hydrochloride (DPZ) and human serum albumin (HSA) were investigated by spectroscopic methods. The association constants, number of binding sites and binding distance of DPZ binding to HSA were enhanced but to different extent in the presence of Cu^2+ , Zn^2+ and F3^3+. The enhancement of the association constants indicated that the binding affinity of DPZ to HSA were enhanced by Cu^2+ ,Zn^2+ and Fe^3+, which will reduce the maximum effect of DPZ and prolong retention time. The conformation of HSA had no change in the presence of Cu^2+ ,Zn^2+ and Fe^3+,which indicated that the higher stability of DPZ-HSA-metal ion complex than DPZ-HSA complex induced the increase of the binding affinity of DPZ to HSA.
