摘 要:目的探讨miRNAhl和miRNA-21在脂联素(ADP)后处理对大鼠心肌缺血-再灌注损伤(MIRI)保护过程中的变化和意义。方法将SD大鼠随机分为假手术组(Sham组)、MIRI组和ADP后处理组(ADP组),每组10只。用Elisa检测各组大鼠血浆心肌肌钙蛋白I(cTnI)水平,用Westernblot检测各组大鼠心肌磷酸化/蛋白激酶B(p-Akt)的表达,用realtimeRT-PCR检测各组大鼠心肌中miRNA_1和miRNA-21的表达。结果与Sham组比较,MIRI组大鼠血浆cTnI水平和心肌miRNA-l表达明显升高,心肌p-Akt和miRNA一21表达明显降低,差异均有统计学意义(P〈0.05);与MIRI组比较,ADP组大鼠血浆cTnI水平和心肌miRNA-1表达明显降低,心肌P-Akt和miRNA-21表达明显升高,差异均有统计学意义(P〈O.05)。心肌miRNA-1表达与血浆cTnI水平呈正相关(r=0.703;P〈0.05),与心肌P—Akt表达呈负相关(r=-0796;P〈0.05);心肌miRNA-21表达与血浆cTnI水平呈负相关(r=-0.727;Pd0.05),与心肌p—Akt表达呈正相关(r=0.886;P〈0.05)。结论miRNA_l是大鼠MIRI及ADP后处理过程中的负性调节因子,miRNA-21是MIRI及ADP后处理过程中的正性调节因子,p-Akt蛋白可能参与了上述调节过程。Objective To observe the changes of two micro-RNA(miRNA-1 and miRNA-21) during adiponectin(ADP) postcon- ditioning against myocardial ischemia reperfusion injury(MIRI) in rats, to study the possible implications of the changes above. Methods SD rats were divided randomly into 3 groups(n=10):(1)Group Sham(group in sham operated);(2)Group MIRI(group in myocardial ischemia reperfusion injury) ;(3)Group ADP(group in ADP postconditioning) were subjected to injection of ADP be- fore reperfusion. The ti.ter of cardiac troponin I(cTnI) in plasma and the the expressions of p-Akt protein in myocardial tissue were observed,the relative expressions of miRNA-1 and miRNA-21 in myocardial tissue in each group were determined by RT-PCR re- spectively. Results Compared with Group Sham,cTnI in plasma and miRNA-1 in myocardial tissue in Group MIRI increased(P〈0.05), p-Akt and miRNA-21 in myocardial tissue declined (P〈0.05) ;compared with Group MIRI, cTnI in plasma and miRNA-1 in myocardial tissue in Group ADP decreased (P^0.05), p-Akt and miRNA-21 in Group ADP increased(P〈0.05), miRNA-1 corre- lated positively with cTnI in plasma and negatively with p-Akt in myocardial tissue(P〈0.05) ;miRNA-21 correlated negatively with cTnI in plasma and positively with p-Akt in myocardial tissue(P〈0.05). Conclusion In the process of MIRI and ADP post- conditioning against MIRI in rats,miRNA-1 is a negative regulatory factor while miRNA-21 is a positive regulatory factor. The reg- ulation above is perherps related to p-Akt.
