摘 要:目的:建立HPLC法测定鱼肝油软膏中全反式维生素A醋酸酯的含量。方法:采用超声提取方法处理鱼肝油软膏,用HPLC法进行含量测定。色谱柱为Platisil ODS C_(18)柱(250 mm×4.6 mm,5μm),流动相:甲醇-水(98:2),流速:1.0 m L·min(-1),检测波长:328 nm,柱温:30℃。结果:维生素A在0.587 9~7.348 5 U·m L^(-1)范围内线性关系良好(r=0.999 9);样品平均回收率为99.78%(RSD=1.57%)。结论:本方法简便、准确、重复性好,可用于鱼肝油软膏中全反式维生素A醋酸酯的含量测定。Objective: To establish the HPLC method for determination of all-trans Vitamin A acetate in cod liver oil ointment. Methods: The Vitamin A acetate in ointment was extracted by ultrasonic method, and the content of all-trans Vitamin A acetate was determinated by HPLC method. HPLC was carried out on a column of Platisil ODS C18 (250 mm × 4.6 mm, 5 μm) with mobile phase consisting of methanol-water (98 : 2). The fow rate was 1.0 mL·min^-1. The detection wavelength was set at 328 nm. The column temperature was 30 ℃. Results: Vitamin A showed a good linear relationship in the range of 0.587 9 – 7.348 5 U·mL^-1 (r = 0.999 9). The average recovery was 99.78% (RSD = 1.57%). Conclusion: The method is simple, accurate and repeatable, which is suitable for the content determination of all-trans Vitamin A acetate in cod liver oil ointment.
