养肝解毒散结中药对肝癌血管生成作用及机制

摘　　要：目的:观察养肝解毒散结(YGJDSJ)中药对肝癌血管生成的作用。方法:裸小鼠皮下移植人肝癌Bel-7402细胞,随杌分为两组,每组10只,分别给予蒸馏水或YGJDSJ灌胃,1次/d,治疗3周后,剥离肿瘤组织,免疫组织化学法检测血管生成及相关基因表达。结果:YGJDSJ中药可以抑制Bel-7402人肝癌组织血管生成,与对照组比较差异有显著性意义(P<0.01);YGJDSJ中药可降低肝癌组织血管内皮生长因子(VEGF)和乏氧诱导因子-1α(HIF-1α)表达,与对照组比较差异有显著性意义(P<0.01);YGJDSJ中药还可降低M2型肿瘤相关巨噬细胞在肝癌组织的分布,与对照组比较差异有显著性意义(P<0.01)。结论:YGJDSJ中药可以抑制肝癌血管生成,其机制与下调VEGF、HIF-1α和M2型肿瘤相关巨噬细胞相关。Objective：To observe the effects of YangGan JieDu SanJie herbs （YGJDSJ） on angiogenesis in human hepatocellular carci- noma. Methods： Nude mice subcutaneously transplanted with Be1 -7402 human hepatocarcinoma ceils were randomly divided into two groups （ 10 mice per group）, and intragastric administered with distilled water or YGJDSJ daily. After three weeks of treatment, the tumors were dis- sected. Gene expression was detected by IHC （Immunohistochemistry）. Results：compared with control group , YGJDSJ could significantly inhibit angiogenesis and down -regulated VEGF （Vascular endothelial growth factor） and HIF-1α （Hypoxia Inducible Factor-1α） expression in Bel-7402 hepatocarcinoma （P 〈 0.01 ）. At the same time,YGJDSJ also could significantly decrease distribution of TAMs （Tumor- associ- ated macrophages） in Be1- 7402 hepatocarcinoma （P 〈 0.01 ）. Conclusion： YGJDSJ can inhibit angiogenesis in hepatocarcinoma and may relate to down-egulation of VEGF, HIF-1α and TAMs.
