摘  要:目的 分析测定煤矿作业工人及尘肺患者呼吸道诱导痰细胞内mRNA表达量,探讨痰细胞内mRNA在早期尘肺诊断中的意义.方法 运用呼吸道诱导痰技术收集113名非尘肺工人、22例尘肺患者及10名健康对照者的痰液,收集痰细胞,运用荧光定量PCR技术测定细胞内转化生长因子1(TGF-1)、表皮生长因子(EGF)、肿瘤坏死因子-α(TNF-α)、白细胞介素-1α(IL-1α)、IL-1、IL-6、IL-8及人成纤维细胞生长因子2(FGF2)的表达量.结果 接尘工龄≥10年组目的基因TGF-1、IL-1和IL-6的mRNA表达量分别是接尘工龄1-2年组的1.40、1.49、1.59倍,其他目的基因表达量比较差异未见统计学意义.其余各组与接尘工龄1-2年组各目的基因mRNA表达量比较差异未见统计学意义(P〉0.05);Ⅰ期尘肺患者组目的基因TGF-1和IL-1的mRNA表达量分别是对照组的1.48、1.53倍,Ⅱ期及Ⅲ期尘肺患者组目的基因TGF-1和IL-1的mRNA表达量分别是对照组的1.49、1.52倍,其他目的基因mRNA表达量比较差异未见统计学意义(P〉0.05).结论 接尘工人和尘肺患者的痰细胞内mRNA表达量的检测,可能对早期检测尘肺有重要意义.Objective To test the expression of mRNA in sputum cells induced from dust-exposed workers and pneumoconiosis patients, in order to investigate the significance of mRNA in sputum cells in early pneumoconiosis diagnosis.Methods Sputum cells of 113 dust-exposed workers, 22 pneumoconiosis patients and 10 healthy people(control group) was sellected by the method of induced sputum.The expression of TGF-1, TNF-α, IL-1α, IL-1, IL-6, IL-8 and FGF2 in sputum cells were detected by reverse transcription-polymerase chain reaction(RT-PCR).Results The mRNA expression levels of TGF-1,IL-1 and IL-6 gene in dust-exposed workers for more than 10 years were 1.40, 1.49, 1.59 times higher than those in dust-exposed workers for more than 1 years.Other target gene was not significantly different between the two groups(P〉0.05).The mRNA expression levels of TGF-1 and IL-1 in stageⅠpneumoconiosis patients were 1.48, 1.53 times higher than those in control group;the mRNA expression levels of TGF-1 and IL-1 in stageⅡand Ⅲ pneumoconiosis patients were 1.49, 1.52 times higher than those in control group;other target gene was not significantly different between the two groups(P〉0.05).Conclusions mRNA detection in sputum cells induced from dust-exposed workers and pneumoconiosis patients may contribute significantly to early diagnosis of pneumoconiosis.
