摘  要:【目的】研究宫内生长受限(intrauterine growth restriction,IUGR)大鼠前额叶皮质内Cofilin、CREB和P38蛋白及其磷酸化表达状况。【方法】通过孕鼠子宫动脉结扎方法建立宫内生长受限大鼠模型,对30 d大鼠进行新位置/新物体实验和Y-型迷宫测试以反映其学习记忆能力。应用Western blotting方法检测30 d大鼠前额叶皮质中Cofilin、CREB、磷酸化CREB、P38及磷酸化P38的表达变化。【结果】IUGR大鼠探索新事物和新空间的时间百分数均明显低于对照组(P<0.05),表明IUGR大鼠学习记忆能力较弱。Western blotting检测显示,IUGR大鼠前额叶皮质中Cofilin蛋白相对表达量与对照组相比明显降低;CREB和P38蛋白相对表达量与对照组相比未见明显差异,但p-CREB明显低于对照组(P<0.01),而p-P38明显高于对照组(P<0.05)。【结论】IUGR大鼠学习记忆功能障碍与Cofilin蛋白的相对表达量下降以及CREB和P38的磷酸化修饰异常有关。[Objective] To study the protein and phosphorylation expressions of cofilin, CREB and P38 in the prefrontal cortex of rats with intrauterine growth restriction (IUGR). [Methods] The rat models of IUGR were established by the ligation of uterine arteries in pregnant rats. On the 30th day, both mine field test and Y-type maze were performed, which reflected the ability of learning and memory. Western blotting was applied to examine the expressions of cofilin, CREB, p-CREB, P38 and p-P38 in the tat prefrontal cortex. [ Results ] Compared with the control group, the time of exploring the new object and space decreased significantly in IUGR group (P〈0.05). It indicated that the ability of learning and memory in IUGR group was poor. Furthermore, the results of western blotting revealed that in the prefrontal cortex of IUGR rats, the relative protein expression of cofllin was significantly lower than that in the control group; there was no obvious difference in the relative protein expressions of CREB and P38 between IUGR group and control group, while the expression of p-CREB was significantly lower than that in the control group (/9〈0.01) and the expression of p-P38 was obviously higher than that in the control group (P〈O.05). [ Conclusion ] The learning and memory dysfunction in IUGR rats is related to the decreasing relative expression of cofilin and abnormal phosphorylation of CREB and P38.
