乙肝后肝硬化中医证型与白介素-28B基因多态性的相关性研究

摘　　要：【目的】探讨乙肝后肝硬化患者白介素-28B(interleukin-28B,IL-28B)基因多态性与乙肝后肝硬化中医辨证分型的关系。【方法】对116例乙肝后肝硬化患者进行中医辨证分型,以118例慢性乙型肝炎患者为对照组,运用基质辅助激光解析电离飞行时间质谱(MALDI-TOF-MS)检测IL-28B基因rs12979860和rs8099917位点的多态性并进行基因分型,分析其基因型与乙肝后肝硬化中医证型之间的关系。【结果】IL-28B基因rs8099917在观察组中TG/GG的基因型频率高于健康对照组(P<0.05),G等位基因可能为乙肝后肝硬化的易感基因。rs12979860位点的基因型及等位基因在两组间未发现差异。乙肝后肝硬化中医证型与IL-28B基因的两个位点的多态性之间未发现有显著的关联(P>0.05)。【结论】IL-28B基因rs8099917位点多态性与乙肝后肝硬化的遗传易感性有关,但未发现rs12979860和rs8099917多态性与乙肝后肝硬化的中医证型相关。[Objective ] To explore the relationship between single nucleotide polymorphism of interleukin-28B （IL-28B） and syndrome types of traditional Chinese medicine （TCM） in patients with HBV-related hepatic cirrhosis. [Methods] A total of 116 cases with HBV-related hepatic cirrhosis were differentiated with TCM syndrome types and 118 patients with chronic hepatitis B were enrolled in the control group. The polymorphism of rs12979860 and rs8099917 in IL-28B was detected by matrix assisted laser desorption ioniza- tion time-of-flight mass spectrometry （MALDI-TOF-MS）. Then the genotypes were differentiated. The relationship was analyzed be- tween genotypes and TCM syndrome types of HBV-related hepatic cirrhosis. [ Results ] As for rs8099917 of IL-28B gene, the frequen- cy of TG/GG genotype in the observation group was higher than that in the control group （P 〈 0.05）, and G allele might be the susceptible gene of HBV-related hepatic cirrhosis. With regard to rs12979860, the genotypes and alleles showed no difference between the two groups. There was no significant correlation between the polymorphism of rs12979860 and rs8099917 in IL-28B and TCM syndrome types of HBV-related hepatic cirrhosis （P 〉 0.05）. [ Conclusion ] The polymorphism of rs8099917 in IL-28B gene may be associated with genetic susceptibility of HBV-related hepatic cirrhosis. However, the polymorphism of rs 12979860 and rs8099917 may not be re- lated with TCM syndrome types of HBV-related hepatic cirrhosis.
