摘 要:目的观察高乌甲素(LA)对脊髓损伤(SCI)大鼠脊髓含水量以及脊髓IL-1β、IL-10、TNF-α水平的影响及其神经保护功能。方法 240只雄性SD大鼠按随机数表法分为SCI组,SCI+生理盐水组(saline组)、SCI+LA组(LA组)和SCI+甲强龙组(MP组),每组60只。每组含有时间点为1、3、6、12、24 h。LA组术后腹腔注射LA,4 mg/kg;saline组术后腹腔注射生理盐水,4 mg/kg;MP组术后立即按首次剂量30 mg/kg腹腔注射,1 h后按5.4 mg/(kg·h)给药,计23 h总量,分4次腹腔注射完毕。在各时间点对各组大鼠进行脊髓含水量测定和脊髓IL-1β、IL-10、TNF-α水平检测。结果 SCI发生后各组各时间点的脊髓含水量明显增加,其中在术后24 h,LA组及MP组脊髓含水量与SCI组比较,差异有显著统计学意义(P<0.01)。SCI后各组各时间点的脊髓IL-1β、IL-10、TNF-α水平明显增加;在术后3、6、12、24 h,MP组及LA组的脊髓TNF-α水平低于SCI组,差异有统计学意义(P<0.05);在术后3、6、12、24 h,MP组及LA组脊髓IL-10水平高于SCI组,差异有统计学意义(P<0.05);在术后各时间点,LA组的脊髓IL-1β水平低于SCI组,差异有统计学意义(P<0.05);在术后6、12、24 h,MP组脊髓IL-1β水平低于SCI组,差异有统计学意义(P<0.05)。结论 LA及MP可减轻SCI大鼠的脊髓水肿程度,降低脊髓IL-1β、TNF-α水平,升高IL-10水平,对SCI大鼠具有神经保护作用。Objective To observe the effect of lappaconitine (LA) on the water content of spinal cord and the spinal cord levels of IL-1β, IL-10 and TNF-α in rats with spinal cord injury (SCI) as well as its neuroprotective function.Methods A total of 240 male Sprague-Dawley rats were randomly divided into SCI group, SCI + saline group (saline group), SCI + LA group (LA group) and SCI + Methylprednisolone group (MP group).With 60 rats in each group, each group contained time points 1, 3, 6, 12, 24 h.The rats in the LA group were injected intraperitoneally immediately with LA (4 mg/kg);the rats in the saline group received intraperitoneal injection of normal saline, 4 mg/kg;the rats in the MP group were injected intraperitoneally immediately with the first dose of 30 mg/kg MP, then were administered according to 5.4 mg/ (kg·h), total amount of 23 h, divided into four injections.The water content of spinal cord and the levels of IL-1β, IL-10 and TNF-α in the spinal cord were measured at each time point.Results Spinal cord water content was significantly increased after SCI.The water content of the spinal cord in LA group and MP group was significantly lower than that in SCI group at the 24 h after SC injury (P〈0.01).After SCI injury, the IL-1β, IL-10 and TNF-α levels in the spinal cord increased significantly in each group.At 3, 6, 12, 24 h after SCI, the TNF-α levels of the spinal cord in LA group and MP group were significantly lower than SCI group (P〈0.05).At 3, 6, 12, 24 h after SCI, the IL-10 levels in the spinal cord in LA group and MP group were significantly higher than SCI group (P〈0.05).The IL-1β levels of spinal cord in the LA group were significantly lower than SCI group at each time point after operation (P〈0.05).The IL-1β levels of spinal cord in the MP group were significantly lower than that in SCI group at 6, 12, 24 h after operation (P〈0.05).Conclusion LA and MP can attenuate the water content of spinal cord after SCI injury, decrease t
