摘 要:目的:观察酚红溶液对栀子苷、小檗碱、黄芩苷含量测定的影响。方法:分别采用紫外分光光度法、酶标仪法、高效液相色谱法检测含有酚红内标物的栀子苷、小檗碱、黄芩苷的含量,并进行方法学考查。结果:当酚红浓度在0.625~40mg/L的范围内,栀子苷、小檗碱、黄芩苷的高效液相法得出较好的线性关系,建立了完整的高效液相方法学。结论:相对于紫外分光光度法和酶标仪法,高效液相法线性好,精确度高,检测方法更为简单可靠,为黄连解毒汤的肠吸收研究提供了高效的检测方法。Objectives To optimize the best detection method of Geniposide,berberine,baicalin in situ single pass intestinal perfusion. Methods Determining the content of geniposide,berberine,baicalin with phenol red as the internal standard,respectively adopting ultraviolet spectroscopy,spectrum microplate spectrophotometer,high performance liquid chromatography. And the methodology of experiment results was investigated. Results Geniposide,berberine,baicalin had a good linear relationship in the range of 0.625 -40mg/L of phenol red. And a complete methodology of high performance liquid chromatography was established. Conclusions The methods of high performance liquid chromatography has a better linear relationship,higher precision,simpler,more reliable than ultraviolet spectroscopy and spectrum microplate spectrophotometer. It provides a more efficient detection method for the study of the intestinal absorption of huanglian jiedu decoction.
