摘  要:目的研究Hedgehog信号通路中分子Shh及Gli-1蛋白在人增生性瘢痕和正常皮肤成纤维细胞中的表达,探讨此通路在增生性瘢痕形成中的作用和意义。方法应用免疫蛋白印记法(western blot)及免疫细胞化学检测15例人增生性瘢痕和正常皮肤成纤维细胞中Shh、Gli-1蛋白的表达情况。结果(1)Western blot检测结果显示,Shh和Gli-1蛋白在增生性瘢痕及正常皮肤成纤维细胞中均有表达,二者在增生性瘢痕成纤维细胞中的表达量(0.640±0.093和0.560±0.120)均比正常成纤维细胞的表达量(0.330±0.023和0.340±0.050)高,其差异具有统计学意义(P<0.05);(2)免疫细胞化学染色结果显示,Shh及Gli-1蛋白在增生性瘢痕及正常皮肤成纤维细胞均有阳性表达,但在增生性瘢痕成纤维细胞组织中的表达明显高于止常皮肤成纤维细胞,其差异均有统计学意义(P<0.05)。结论增生性瘢痕成纤维细胞中,Shh及Gli-1蛋白呈现高表达,提示Shh信号通路异常激活可能在增生性瘢痕发生过程中起到了重要作用。Objective To investigate the expression of Shh and Gli-1 in the Sonic hedgehog signaling pathway in human hypertrophic scar and normal skin fibroblasts in vitro,and to explore the significance of this signaling pathway in hypertrophic scar formation.Methods The expression of Shh and Gli-1 in 15 samples of human hypertrophic scar and normal skin fibroblasts was detected by western blot and immunocytochemistry.Results (1) Western blot showed that both normal human skin and hypertrophic scar fibroblasts had Shh and Gli-1 protein expression,and their expressions in the hypertrophic scar fibroblasts [(0.640 ± 0.093) and (0.560 ± 0.120)] were higher than that in the normal human skin fibroblasts [(0.330 ± 0.023) and (0.340 ± 0.050)].The difference was statistically significant (P < 0.05).(2) Immunocytochemistry showed that both Shh and Gli-1 were expressed in both hypertrophic scar and normal skin fibroblasts.The level of Shh and Gli-1 protein was significantly raised.Conclusion The expression of Shh and Gli-1was elevated in hypertrophic scar fibroblasts which indicated that the Sonic hedgehog signaling pathway may play an important role in the occurrence of hypertrophic scars.
