摘  要:目的进行钩藤饮片生物碱组分(AUR)制备,探索AUR对D-半乳糖模型AD大鼠学习记忆功能作用。方法单因素筛选大孔吸附树脂制备钩藤饮片生物碱部位方法。采用D-半乳糖腹腔注射制备AD大鼠模型,采用Morris水迷宫测试大鼠的学习和记忆能力,采用分光光度法测定血清中乙酰胆碱转移酶(ChAT)、乙酰胆碱酯酶(Ach E)、乙酰胆碱(Ach)、超氧化物歧化酶(SOD)、丙二醛(MDA)活性。结果选择75%乙醇作为树脂洗脱溶媒。AUR能够增加AD模型大鼠的入水象限停留时间,缩短逃避潜伏期,提高血清ChAT、Ach、SOD水平,降低Ach E活性、MDA水平。结论 AB-8树脂适用于AUR制备,AD模型大鼠的学习记忆功能降低,AUR可改善AD大鼠的学习记忆能力,其机制与改善胆碱能系统作用有关。Objective To study the effect of alkaloids of U. R.( AUR) on memory,Ach E and antixidation ability in Alzheimer's disease( AD) rat model. Methods With the yield of total alkaloids as a marker,the static and dynamic adsorption/desorption methods were used to investigate the optimum process factors by the single factor experiment. The cognitive impairment rat model was injected by D-galactose. The learning and memory ability was determined by Morris water maze test. The level of SOD,MDA,Ach,Ch AT and Ach E were measured by spectrophotometer. Results Compared with control group rats,AD model rats' learning and memory ability was severely impaired,SOD,Ach,Ch AT level significantly decreased,MDA and Ach E level increased. The administration of AUR improved memory ability,improved rats' SOD,Ach and Ch AT,reduced MDA and Ach E activity in serum. Conclusion AUR can improve learning,memory,spatial cognitive ability and antioxidant stress ability,increase SOD,Ach,ch AT level of AD rats.
