摘  要:[目的]探究甘草次酸(GA)影响前列腺癌(PC3)细胞BLM基因的转录以及细胞的增殖、迁移、侵袭和凋亡的情况。[方法]MTT法检测细胞增殖能力并筛选适宜的药物浓度,荧光定量PCR检测BLM基因的表达量,划痕法检测细胞迁移,Transwell小室检测细胞侵袭,流式细胞仪检测细胞凋亡。[结果]甘草次酸浓度在0.12 mol/L^0.36 mol/L范围时,随着浓度增大,细胞的增殖能力逐渐下降,呈现药物浓度梯度依赖性;当甘草次酸浓度为0.2 mol/L时,PC3细胞中BLM的mRNA的表达量仅有对照组的0.2倍;甘草次酸浓度在0.10 mol/L以上时,细胞的侵袭和迁移能力均显著减小;流式结果显示甘草次酸浓度在0.10 mol/L时,细胞凋亡率为45.25±0.5**,差异极显著(P<0.01)。[结论]甘草次酸浓度为0.15 mol/L以上时,能够下调PC3细胞中BLM基因的转录量(P<0.01),在0.12 mol/L^0.36 mol/L范围时,对PC3细胞的增殖、侵袭、迁移具有抑制作用,对PC3细胞凋亡具有促进作用。[ Objective ] In the current,the expression of BLM was detected and investigated the effect of glycyrrhetinic acid (GA) on transcription, cell proliferation, migration, invasion and apoptosis of prostate cancer ( PC3 ) cells. [ Methods ] Cell proliferation and drug screening of the suitable concentration had been evaluated by the MTT. The expression of BLM gene was detected by RT - qPCR, cell scratch was used to examine cell migration. Cell invasion was detected by Transwell and cell apop- tosis was assessed by flow cytometry. [ Results] The concentration of glycyrrhetic acid in 0.12 mol/L- 0.36 mol/L range ,with the increase of concentration, cell proliferation gradually decreased with concentration dependent. The concentration of glycyr- rhetic acid is 0.2 mol/L,0.2 times the expression level of PC3 cells in BLM mRNA was less than the control group. The con- centration of glycyrrhetic acid in the above 0.10mol/L, the migration and invasion of cells had been significantly reduced. Flow cytometry showed that the apoptotic rate was 45.25 ± 0.5 * * when the concentration of glycyrrhetinic acid was 0.10mol/L, the difference was significant (P 〈 0.01 ). [ Conclusion] The proliferation, invasion and migration of PC3 cells were suppressed and promoted the apoptosis of PC3 cells when the concentration of glycyrrhetinic was acid in the range of 0.12 moL/L to 0.36 mol/L in PC3 cells ,when the concentration of glycyrrhetinie acid was 0.15 mol/L or more can down -regulated the BILM gene transcription.
