摘 要:长非编码RNA(lncRNA)是重要基因调节者,参与多种疾病的发生。本研究着重测定了填饲和脂肪肝相关因子对lncRNA基因(LOC106047490)表达的影响。在填饲7、14、19 d时,该基因在填饲鹅肝脏和腹脂中的表达均受到不同程度的抑制,而在胸肌仅第19天时有较明显的抑制。高葡萄糖、胰岛素和不饱和脂肪酸可在鹅原代肝细胞中诱导该基因的表达,但饱和脂肪酸无诱导作用。此外,在常规饲料中添加20%的蔗糖虽可提高鹅肥肝产量,但并不诱导该基因的表达。最后,本研究还发现该基因在鹅胚胎的肠和肌胃中表达最高,在肝和腿肌中表达最低。综上所述,LOC106047490在鹅肥肝形成过程中扮演重要角色,其表达受葡萄糖、胰岛素和不饱和脂肪酸影响,这为研究其功能提供了良好的体外模型。Long non-coding RNAs( lncRNAs),as key player in gene regulation,are involved in the occurrence of several diseases. This study investigated the effect of overfeeding and fatty liver associated factors on expression of goose lncRNA,LOC106047490. The expression of the gene was inhibited to different degree in both the liver and abdominal fat of the overfed geese at day 7,day 14 and day 19 post-overfeeding,while in breast muscle the inhibition became apparent only at day 19 post-overfeeding. High glucose,unsaturated fatty acids and insulin could induce its expression,but saturated fatty acid did not. In addition,supplementation of 20% suger to the normal diet could increase the average weight of fatty liver in the overfed geese,but did not induce the expression of the gene. Finally,the expression of LOC106047490 was found high in embryonic intestine and gizzard,but low in liver and thigh muscle. In conclusion,LOC106047490 may play an important role in the development of goose fatty liver,and its expression was regulated by glucose,insulin and unsaturated fatty acids,which provides an in vitro model for studying the function of the lncRNA gene.
