摘 要:伴随高通量测序技术的快速发展,转录组测序(RNA-Seq)技术在各个研究领域已经得到广泛的运用.RNA-Seq通过分析不同细胞或者组织转录组的表达情况来揭示细胞的基因表达情况,结构特点和调控规律.近年来基于高通量测序技术的RNA-Seq分析方法发展迅速,涌现出一大批相关的分析方法和工具,如何根据实际需求选择合适的工具和分析流程,成为广大科研人员面临的问题.本文参照近几年在RNA-seq技术研究领域发表的文献,综述了RNA-Seq应用实际过程中涉及的分析方法、软件工具及其选用标准,为相关的研究和应用提供信息和参考.With the remarkable advances of high-throughput sequencing technology, transcriptome sequencing (RNA-Seq) technology has been widely used in various research fields.RNA-Seq can disclose gene expression spectrum, structure feature and regulation network in live cells by differential RNA-Seq on whole cells in different conditions or from distinct tissues.While the rapid application of high-throughput sequencing technology,there are a large number of emerging analytical methods and toolsfor RNA-Seq, which sometimes puzzles the researchers how to make a choice on an appropriate algorithm analysis and corresponding tool.Here, the analytical methods, software tools and selection criteria for practical RNA-Seq analysiswere reviewed in the literature, and then provide information and insights for related research and application.
