扶正消癥方对环磷酰胺治疗S180荷瘤小鼠的增效减毒作用

摘　　要：目的观察扶正消癥方对环磷酰胺治疗S180荷瘤小鼠的增效减毒作用。方法将腋下接种S180的小鼠随机分为肿瘤组,环磷酰胺组(0.02 g/kg)、环磷酰胺+扶正消癥方[(0.02+1.43)g/kg和(0.02+0.36)g/kg]组,另设正常对照组。除肿瘤组及正常对照组外,其他各组于接种24 h后腹腔注射环磷酰胺,环磷酰胺+扶正消癥方组按剂量灌胃扶正消癥方溶液,正常对照组、肿瘤对照组灌胃等量纯净水,每天1次,连续10 d。实验结束进行血细胞计数,统计抑瘤率、脾脏、胸腺及肝脏脏器指数,生化法检测血清中谷丙转氨酶(ALT)、谷草转氨酶(AST)、血肌酐(SCr)的含量;Elisa法检测肿瘤匀浆中肿瘤特异性坏死因子(TSGF)、磷酸化蛋白激酶C(P-PKC)、端粒酶(TF)含量。结果与环磷酰胺组比较,扶正消癥方联合给药组可明显升高S180荷瘤小鼠化疗后血细胞数量,能显著提高抑瘤率并且增加免疫器官重量,减少血清中GOT、GPT的含量,减少肿瘤匀浆中TSGF、P-PKC、TE的含量(P<0.05,P<0.01),并且以1.43 g/kg效果最佳。结论扶正消癥方对环磷酰胺抑制肿瘤有良好的增效作用,并且能够减轻环磷酰胺对机体的损伤,体现出一定的增效减毒作用。Objective To observe the effect ofFuzheng Xiaozheng decoction on efficacy enhancing and toxicity reducing in combination with cyclophosphamide （CTX） in mice transplanted S180 tumor.Methods The mice were randomly divided into two groups inoculated with S180, respectively. The tumor group, cyclophosphamide group （20 mg/kg）, cyclophosphamide and Fuzheng Xiaozheng decoction [（0.02＋1.43） g/（kg＆#183;d）and（0.02＋0.36） g/（kg＆#183;d）] group, and normal control group. In addition to tumor group and the normal control group, the other groups in 24 h after inoculation with intraperitoneal injection of cyclophosphamide, the normal control group and the tumor control group under the same conditions were fed with the same amount of pure water, 1 times a day, continuous 10 d. After the experiment, blood cell count was performed; Tumor suppression rate was calculated; The spleen, thymus and liver weight, serum ALT and AST SCr content usin g ELISA method to detect TSGF, P-PKC, TF content in tumor tissue. ResultsFuzheng Xiaozheng decoction has efficacy inhibit on transplanted tumor S180, and can enhance white blood cells, immune organ index, and has good protective effects on liver and kidney damage caused by cyclophosphamide and reduce the content of TSGF, P-PKC, TE in tumor tissue. （P〈0.05,P〈0.01）, and the best effect of 1.43 g/kg.Conclusion This study showing theFuzheng Xiaozheng decoction can synergy efficacy enhancing and toxicity reducing in combination with cyclophosphamide （CTX） in mouse transplanted S180 tumor.
