摘 要:目的了解云南大理某农村猪群中星状病毒(astrovirus,AstV)感染情况,为AstV的防控提供理论依据。方法利用逆转录巢式聚合酶链反应(RT-nested PCR)技术,对云南大理某农村采集的42份猪粪便样品进行PAstV799 bp基因部分片段扩增、克隆,并测序;利用NCBI BLAST进行数据库比对,Mega 4.1软件进行序列同源性比较和系统进化树构建。结果 Trizol试剂提取42份猪粪便样品上清总RNA,经RT-nested PCR扩增,5份样品可见799 bp的目的基因片段,阳性率为11.90%;PAstV 799 bp片段同源性分析显示,5个毒株与PAstV-1、2、3、4和5的同源性为46.3%~87.9%,与PAstV-4的同源性最高,为75.5%~87.9%;PAstV 799 bp片段的系统进化分析显示,分离的毒株属于PAstV-4,同源性为80.0%~99.5%。结论云南省PAstV-4流行病学调查将有利于中国AstV的预防及治疗。Objective To investigate the prevalence of porcine astrovirus(Ast V) infection in rural area of Dali City,Yunnan Province, China, and provide a theoretical basis for the prevention and control of Ast V. Methods A 799 bp PAst V gene fragment was amplified from 42 swine stool samples by reverse transcription-nested polymerase chain reaction(RT-nested PCR), then cloned, sequenced, searched in NCBI BLAST data, and analyzed for homology by Mega 4. 1software, based on which a phylogenetic tree was constructed. Results Total RNA was extracted from the supernatant of42 swine stool samples by using Trizol reagent, with 5 of which the target gene was amplified by RT-nested PCR,indicating a positive rate of 11. 90%. The homologies of five strains to PAst V-1, 2, 3, 4 and 5 were 46. 3% ~ 87. 9%,while those to PAst V-4 were the highest, which were 75. 5% ~ 87. 9%. Phylogenetic analysis showed that the isolated strains belonged to PAst V-4, of which the homologies were 80. 0% ~ 99. 5%. Conclusion The epidemiological investigation on PAst V-4 in Yunnan Province will be beneficial to the prevention and treatment of Ast V in China.
