摘 要:目的:探讨人脐带间质干细胞来源的外泌体(exosome from human umbilical cord mesenchymal stem cells,hucMSC-ex)对顺铂抗肿瘤效果的影响,评价hucMSC-ex的安全性。方法:蛋白质印迹法检测外泌体的表面特异性分子CD9、CD63以鉴定hucMSC-ex。通过细胞抑制率测定、克隆形成实验、细胞迁移实验,检测hucMSC-ex存在条件下顺铂对HGC-27细胞和MGC-803细胞抑制率、克隆形成及迁移效率的影响。结果:HucMSC-ex表达外泌体特异性分子标志物CD9、CD63;hucMSC-ex能减弱顺铂对HGC-27细胞和MGC-803细胞的生长抑制作用,但不影响顺铂作用后的HGC-27细胞和MGC-803细胞克隆形成及迁移效率。结论:hucMSC-ex对顺铂抗肿瘤疗效无明显影响,具有一定的安全性。Objective:To explore the effects of exosome from human umbilical cord mesenchymal stem cells(hucMSC-ex) on cisplatin chemotherapy and to assess the safety of hucMSC-ex.Methods:The specific molecular marker of exosome were detected by Western blotting.By detecting the inhibitor rate, clone formation and migration efficiency of cisplatin to HGC-27 and MGC-803 with treatment of hucMSC-ex to explore the effects of hucMSC-ex.Results:HucMSC-ex expressed the specific marker CD9 and CD63.HucMSC-ex could affect the inhibitor rate of cisplatin to HGC-27 and MGC-803 cells slightly.However, hucMSC-ex did not influence the the clone formation and migration efficiency of cisplatin to HGC-27 and MGC-803 cells.Conclusion:Exosome from human umbilical cord mesenchymal stem cells exerted minor influence on cisplatin chemotherapy and were relatively safe.
