摘　　要：目的探讨NLRP3炎性体信号通路在急性痛风性关节炎患者中的变化。方法选取来我院就诊的360例患者作为研究对象。设置3个实验,分别为蛋白质免疫印迹实验、酶联免疫吸附测定实验和关节滑膜免疫组化实验。每个实验设6组对照实验(正常组,模型组,秋水仙碱阳性药组及GT低、中、高剂量组),每组20例患者。取样观察患者关节滑膜组织,检测白细胞介素-6(IL-6),肿瘤坏死因子-α(TNF-α)及Nod样受体蛋白3(NLRP3)炎性体的表达,分析测定平均积分吸光度(IA)。结果治疗前后对比发现GT中、高剂量组的NLRP3的表达水平降低(P<0.05),GT各组IL-1β、IL-6、TNF-α、NF-κB表达均明显降低(P<0.05)。结论使用GT和秋水仙碱治疗急性痛风性关节炎时可能会降低NLRP3炎性体信号通路中炎性因子的表达,对患者体内的NF-κB的活化和IL-1β分化成熟也会有一定的影响。Objective To investigate the changes of NLRP3 inflammatory signaling pathway in patients with acute gouty arthritis. Methods A total of 360 patients admitted in our hospital were selected as the research objects. Three experiments were performed, including protein immunoblotting experiment, enzyme-linked immunosorbent assay and synovial immunohistochemistry experiment. In each experiment, six groups of control experiments（normal group, model group, colchicine positive drug group and GT low, medium and high close group） were set,with 20 patients in each group. Sam ples in synovial tissues were observed. The expressions of interleukin-6（IL-6）, tumor necrosis factor-ot（TNF-α） and Nod-like receptor protein 3（NLRP3） were detected. The average integral absorbance （IA） was measured. Results It was found that the expression levels of NLRP3 in the GT middle and high dose groups were decreased after treatment, compared with those before treatment （P〈0.05）. The expressions of 1β, IL-6, TNF-α and NF-κB were significantly decreased in the GT groups（P〈0.05）. Conclusion The use of GT and colchicine in the treatment of acute gouty arthritis may reduce the expressions of inflammatory cytokines in the NLRP3 signaling pathway, and will have a certain impact on the activation of NF-κB and differentiation and maturation of IL-1β in patients.
