The Effects of Flavonoid Aglycones Extracted from LianXing JiDou on Immunologic Function of the Immunosuppressive Mice

摘  要:目的:探讨藏药镰形棘豆黄酮苷元对免疫抑制小鼠细胞和体液免疫功能的影响。方法:将60只昆明种小鼠随机分为6组:正常组、模型组、盐酸左旋咪唑组(25 mg/kg)及黄酮苷元高、中、低剂量组(336,168,84 mg/kg),每组10只。高、中、低剂量组分别灌胃给予不同剂量的黄酮苷元,正常组和模型组灌服等体积5%吐温-80溶液,共10天。第8天开始注射环磷酰胺制备免疫抑制小鼠模型,除正常组腹腔注射生理盐水外,其他各组分别腹腔注射环磷酰胺,共3天。采用ConA诱导的小鼠脾淋巴细胞转化试验、NK细胞活性的测定、血清溶血素的测定方法观察镰形棘豆黄酮苷元对免疫抑制小鼠免疫功能的影响。结果:与模型组比较,镰形棘豆黄酮苷元各剂量组能明显提高免疫抑制小鼠的淋巴细胞转化能力(P<0.05)、NK细胞活性百分率(P<0.05)和血清半数溶血值(HC_((50))(P<0.05)。结论:镰形棘豆黄酮苷元能够提高环磷酰胺造成的免疫抑制小鼠的免疫功能。Objective: To investigate the effects of the flavonoid aglycones extracted from LianXing JiDou (Oxytropis fdcata Bunge) on cellular immunity and humoral immunity of immunosuppressive mice. Methods: Sixty Kunraing mice were randomly divided into six groups: the normal group, the model group, levamisole group (25 mg/kg), high, moderate and low dose groups of flavonoid aglyeones (336, 168, 84 mg/kg), ten mice each group. High, moderate and low dose groups of flavonoid aglycones accepted intragastric administration of flavonoid agly- cones in different concentrations respectively, and the normal group and the model group received the lavage of 5% tween-80 solution, for ten days. The immunosuppressive mice modesl were prepared by injecting cyclophosphamide since the eighth day in other groups for three days except the normal group accepted intraperitoneal injection of physiological saline. The effects of flavonoid aglycones on the immunologic function of mice were observed by using splenic lymphocyte transformation test of the mice induced by concanavalin A (ConA), the determination of NK cell activity, the determination of serum hemolysin. Results: Compared with the model group, different dosages groups offlavonoid aglycones could obviously raise lymphocyte transformation ability of immuno- suppressive mice (P〈0.05), NK cells activity percentages (P〈0.05) and HDs0 values (P〈0.05). Conclusion: Flavonoid aglycones extracted from LianXing JiDou could enhance the immunity of eyclophosphamide-induced im- munosuppressive mice.
