摘 要:目的探讨中药气血宁对心肌梗死大鼠心脏结构、功能及ghrelin表达的影响。方法将雄性SD大鼠随机分为假手术组、模型组、倍他乐克组、气血宁小剂量组和气血宁大剂量组。采用结扎大鼠心脏左冠状动脉前降支的方法制作心肌梗死大鼠模型,假手术组平行手术操作但不结扎前降支。术后灌胃给药4周。采用超声心动图检测心脏结构、功能,HE染色观察心肌组织病理,实时荧光定量PCR检测ghrelin及其受体GHSR mRNA的表达。结果与假手术组比较,模型组、倍他乐克组、气血宁小剂量组和气血宁大剂量组收缩末期的室间隔厚度均显著变薄(P<0.05,P<0.01),左室内径显著增大(P<0.01),左室射血分数和左室短轴缩短分数明显降低(P<0.01);模型组舒张末期的左室内径显著增大(P<0.05),心肌组织ghrelin及其受体GHSR mRNA的表达显著增高(P<0.01)。与模型组比较,气血宁大剂量组左室射血分数和左室短轴缩短分数明显增大(P<0.05);倍他乐克组、气血宁小剂量组和气血宁大剂量组的心肌组织病理损伤有所改善,ghrelin及其受体GHSR mRNA的表达显著降低(P<0.05,P<0.01)。结论中药气血宁能部分改善心肌梗死大鼠心脏结构、功能,调节ghrelin及其受体GHSR mRNA的表达。Objective To study the effects of Chinese herbal medicine Qixuening on cardiac structure, function and ghrelin expression in rats with myocardial infarction. Methods Male SD rats were randomly divided into sham operation group, model group, Betaloc group, Qixuening low dose group and Qixuening high dose group. The rat model of myocardial infarction was produced by tigation of anterior descending branch of left coronary artery, and the sham operation group was operated by parallel operation without ligation. The rats had intragastric administration for 4 weeks. Then the cardiac structure and function weredetected by eehocardiography. The pathological changes of myocardium were observed by HE staining. And ghrelin and its receptor GHSR mRNA expression were detected by real time fluorescent quantitative PCR. Results Compared with the sham operation group, the interventricular septum end-stolic thickness got thinner, the left ventrieular dimension was increased significantly( P 〈 0. 01 ), left ventricular ejection fraction and the left ventricular fractional shortening were decreased significantly ( P 〈 0. 01 ) in the model group, Betaloc group, Qixuening low dose group and Qixuening high dose group; and the left ventrieular end-systolic dimension in the model group increased significantly( P 〈 0. 05 ), cardiac tissue ghrelina and its receptor GHSR mRNA were increased (P 〈 0. 01 ). Compared with the model group, left ventrieular ejection fraction and left ventricular fractional shortening were increased significantly( P 〈 0. 05 ) in Qixuening high dose group. Ghrelin and its receptor GHSR mRNA expression decreased significantly( P 〈 0.05, P 〈 0.01 ), and the pathological damage of myocardial tissue was improved in Betaloc group, Qixuening low dose group and Qixuening high dose group (P 〈 0. 05, P 〈0. 01 ). Conclusion Chinese herbal medicine Qixuening can improve the cardiac structure and function, regulate the expression of ghrelin and its receptor GHSR mRNA in rats with
