摘  要:目的观察保济丸不同剂量^(60)Co辐照对葛根素、橙皮苷、柚皮苷质量分数及对小鼠小肠推进作用的影响。方法采用高效液相色谱法分析保济丸主要成分辐照前后变化,色谱柱为Agilent Zorbax SB-C_(18)(150 mm×4.6 mm,5μm),柱温为25℃,流速为1 mL/min,以甲醇(A)-水(B)为流动相,梯度洗脱(0~20min,30%A;20~30 min,30%→75%A;30~40 min,75%→80%A;40~45 min,80%→30%A),检测波长为250 nm(葛根素)和283 nm(柚皮苷、橙皮苷)。动物实验分组为正常对照组(Normal),保济丸辐照前组(BBJ),保济丸低剂量辐照组2 kGy(BJL)、中剂量辐照组6 kGy(BJM)、高剂量辐照组10 kGy(BJH),和阳性药物阿托品组(ATP),Normal组灌服生理盐水,ATP组灌服3 mg/kg硫酸阿托品溶液,其余组灌胃3 g/kg各相应剂量组保济丸,以小鼠小肠炭末推进试验评价保济丸对胃肠功能的作用。结果葛根素、橙皮苷、柚皮苷分别在8.9~178 mg/L(r=0.999 37,n=6)、7.25~232 mg/L(r=0.999 94,n=6)、2.65~84.8mg/L(r=0.999 95,n=6)范围内呈良好的线性关系;平均回收率分别为100.60%(RSD=3.49%)、102.02%(RSD=4.46%)、103.16%(RSD=3.79%)。不同辐照剂量辐照的保济丸3种成分的质量分数与辐照前比较均无统计学意义(P>0.05)。与正常组相比,不同辐照剂量保济丸都可显著抑制正常小鼠的小肠推进距离和推进率(P<0.05)。结论不同剂量^(60)Co辐照对保济丸主要3种成分质量分数均无显著影响;对正常小鼠小肠推进功能均有抑制作用,建议尽可能选择低剂量辐照灭菌。Objective To investigate the effect of different dosages of Baoji Pills' ^60Co irradiation on contents of puerarin,hesperidin and naringine,and on intestinal propulsion in normal mice. Methods The contents of puerarin, hesperidin and naringine before and after irradiation were analyzed by HPLC, and chromatographic column was Agilent Zorbax SB-C18( 150 mm× 4.6 mm,5 μm). Column temperature was 25℃ and the flow rate was 1 mL/min. Gradient eluting was performed with alcohol( A)-H_2O( B)( 0-20 min,30%A; 20-30 min,30%→75% A; 30-40 min,75%→80% A; 40-45 min,80%→30%A),and the detection wave length was 250 nm for puerarin and 283 nm for hesperidin and naringine. The experimental groups included normal group, the group before irradiation of Baoji pills( BBJ), the low dose irradiation group of Baoji pills( BJL),the medium dose irradiation group of Baoji pills( BJM),the high dose irradiation group of Baoji pills( BJH) and the positive group of atropine( ATP). The rats were administered normal saline in normal group and atropine sulphate 3 mg/kg in positive group,the other groups were administered 3 g/kg different doses of Baoji pills. The gastrointestinal function of Baoji pills were evaluated by carbon medicinalis drive experiment. Results Puerarin,hesperidin and naringine showed good linear relationship within the ranges of 8.9-178 mg/L( r = 0.999 37,n = 6),7.25-232 mg/L( r = 0.999 94,n = 6), and 2.65-84.8 mg/L( r = 0.999 95,n = 6),respectively. The average recovery were 100.60%( RSD = 3.49%),102.02%( RSD = 4. 46%), 103. 16%( RSD = 3. 79%), respectively. There was no significant difference between different irradiation dosages of Baoji pills( P〈0.05). Compared with normal group,intestine boost distance could be inhibited significantly by each dose of Baoji pills( P〈0.05).
