摘 要:目的对大鼠进行苯并[a]芘(B[a]P)急性染毒后用光镜和电镜观察其海马组织的形态学改变,从而探究B[a]P对大鼠海马神经细胞的影响。方法将50只SD大鼠随机分为对照组(n=10)和实验组(n=40),其中实验组又按大鼠染毒后的处死时间分为5组(染毒后1h组、染毒后1d组、染毒后2d组、染毒后3d组、染毒后5d组),每组8只。对实验组大鼠尾静脉注射B[a]P,对照组大鼠尾静脉注射相同剂量的生理盐水。染毒结束后,分别在不同的特定时间点取下大鼠的海马组织在光镜和电镜下进行观察。结果与对照组相比,实验组大鼠B[a]P急性染毒后出现了易受惊、易兴奋、易激惹、感觉迟钝、反应低下等异常表现。各组大鼠海马组织光镜观察结果显示,实验组大鼠海马神经细胞数量及形态与对照组大鼠比较,没有明显变化。而各组大鼠海马组织电镜观察结果显示,实验组大鼠的海马神经细胞出现了胞体浓缩变性、细胞核皱缩、粗面内质网扩张、线粒体肿胀等。结论 B[a]P急性染毒对大鼠海马神经细胞具有损伤作用。Objective To research the effect of Benzo[a]pyrene (BaP) on morphological changes of hippocampal neurons in rats that exposed to BaP through observing the morphological changes of hippocampus tissues BaP by light microscope and electron microscope. Methods 50 SD rats were randomly divided into control group (n = 10) and experimental group (n = 4:0). After being exposed to BaP, the rats in experimental group were divided into 5 groups according to the time they have been executed (1 h after exposure group, 1 d after exposure group, 2 d after exposure group, 3 d after exposure group , 5 d after exposure group), and each group of 8 SD rats. BaP was injected into rats of experimental group via caudal vein, similarly the rats of control group were injected with normal saline of same dosage as experimental group. After the BaP exposure, the hippocampus tissues of rats were obtained for observation by light microscope and electron microscope at specific points in time. Results Compared to the control group, rats in experimental group after BaP exposure were easily to show abnormal symptoms of being surprised, excited, irritated, insensitive and slow in response. Through the observation of light microscope, in the comparison of 2 groups, there was no significant difference between their quantities and forms of hippocampal neuron. And electron microscope results of each ratrs hippocampus showed that there were concentrated degeneration of soma, plasmolysis, expansion of rough endoplasmic reticulum, and mitochondria swelling, etc. Conclusion Acute BaP exposure could damage the hippocampal neurons of rats.
