摘 要:目的探究苯并[a]芘(B[a]P)慢性染毒对SD大鼠学习记忆能力的影响。方法将60只SD大鼠随机分为溶剂对照组及B[a]P染毒组(B[a]P低剂量染毒组、中剂量染毒组和高剂量染毒组),对溶剂对照组大鼠腹腔注射植物油,对染毒组大鼠腹腔注射不同剂量B[a]P,连续注射10周。染毒结束后,采用Morris水迷宫实验对大鼠学习记忆能力进行测试。结果 B[a]P染毒组多数大鼠在研究期间均有异常行为表现。定向导航实验中,各组大鼠的平均逃避潜伏期随着实验天数的增加而缩短,且在同一天内,各组大鼠的平均逃避潜伏期比较,差异均有统计学意义(P<0.01)。其中,溶剂对照组大鼠的平均逃避潜伏期明显短于B[a]P染毒组大鼠,且在实验的前4天,随着染毒剂量的增加,大鼠的平均逃避潜伏期越长,差异均有统计学意义(P<0.05)。在空间探索实验中,各组大鼠第1次找到原平台位置所用时间、穿越原平台位置次数及在原平台象限的停留时间比较,差异均有统计学意义(P<0.01)。其中,溶剂对照组大鼠第1次找到原平台位置所用时间短于B[a]P染毒组大鼠,穿越原平台位置次数多于B[a]P染毒组大鼠,在原平台象限的停留时间长于B[a]P染毒组大鼠,且随着染毒剂量的增加,大鼠第1次找到原平台位置所用时间越长,穿越原平台位置次数越少,在原平台象限的停留时间越短,差异均有统计学意义(P<0.05)。结论 B[a]P慢性染毒对大鼠的学习记忆能力有损害作用,且大鼠的学习记忆能力随B[a]P染毒剂量增加呈逐渐降低趋势,表明B[a]P对大鼠学习记忆能力的损害存在一定的剂量效应关系。Objective To research effects of chronic exposure to benzo[a]pyrene (BaP) on learning and memory ability of SD rats. Methods 60 SD rats were randomly divided into solvent control group and BaP exposure group( low-dose group, medium-dose group and high-dose group). In the solvent control group, vegetable oil was injected into the rats' abdominal cavity, and different doses of gap were injected into rats' abdominal cavi ty in the BaP exposure group for 10 weeks. After the BaP exposure, the Morris maze experiment was used to evaluate learning and memory ability of rats. Results In the process of study all the rats exposed to BaP showed some abnormal behaviors. In the navigation experiment, as the days of experiment increased escaping and hiding time of rats from each group decreased. And within a day the average escape latent period of rats in each group was different with statistical significanre (P 〈 0.01 ). The average escape latent period of solvent control group was obviously shorter than the BaP exposure group, and during 4 days after the experiment started as the dose of BaP exposure increased, the average escape latent period of rats increased correspondingly, and there was statistical significance(P〈0.05).In the space exploration experiment, when compared the time needed to find the original platform the first time, the number of times crossed the original platform and lengthy of stay on the original platform for rats in each group, the differences was statistical significant(P〈0.01), among which the rats in solvent control group used less time to first find the original platform and crossed the original platform more often than BaP exposure group and also stayed longer on the original platfom, and as the dose B[a]P increased, the rats used more time getting back to the original platform first, crossed the original platform less times and stayed shoter on the platform quadrart, and there was statistical significance(P〈0.05).
